Mll partial tandem duplication and Flt3 internal tandem duplication in a double knock-in mouse recapitulates features of counterpart human acute myeloid leukemias

Abstract: The MLL-partial tandem duplication (PTD) associates with high-risk cytogenetically normal acute myeloid leukemia (AML). Concurrent presence of FLT3-internal tandem duplication (ITD) is observed in 25% of patients with MLL-PTD AML. However, mice expressing either Mll-PTD or Flt3-ITD do not develop AML, suggesting that 2 mutations are necessary for the AML phenotype. Thus, we generated a mouse expressing both Mll-PTD and IntroductionAcute myeloid leukemia (AML) is a genetically heterogeneous disease. Recurrent … Show more

“…The ectopic myeloid expansion corroborated with murine studies in which Flt3 knockin induced myeloid expansion reminiscent of human myeloproliferative neoplasms 44 and AML with short latency in the presence of cooperative oncogenic signaling. 45,46 Importantly, coexisting TKD in the same allele conferred the myeloid compartment with the resistance to AC220, consistent with the clonal emergence of AC220-resistant AML patients.…”

“…For multiparameter flow cytometry conducted on murine cells, single-cell suspensions were obtained from spleen, bone marrow, lymph node, or blood as previously described (58,61). Briefly, organs were dissociated manually and washed with RPMI 1640 supplemented with 10% FBS and 0.5% EDTA, and rbc were lysed with ammonium chloride rbc lysis buffer (Stem Cell Technologies).…”

“…24 This model presents clinical, cytogenetic, and molecular features that match the human counterpart, 2,25,26 thereby representing a suitable model to study novel leukemogeneic mechanisms and therapeutic approaches. It is also particularly suitable for testing echinomycin as we have previously shown the drug to be effective in surrogate leukemia stem cell assays with 7 cases of human AML, 3 of which harbor FLT3 mutations.…”
