Modified Benzidine Test for the Detection of Cytochrome-Containing Respiratory Systems in Microorganisms

Abstract: Most aerobic and facultative bacteria contain a cytochrome system that serves as a terminal electron carrier during aerobic respiration. The majority of these bacteria also produce catalase that can decompose any hydrogen peroxide that might be produced by flavoprotein oxidases. These two iron-porphyrin systems generally are not present in anaerobic bacteria or in lactic acid bacteria. This correlation has led to employment of the simple test for catalase as a general test for iron-porphyrin systems, and a neg… Show more

“…Another clue to the anaerobic nature of the mutant was suggested by its negative benzidine test. This test was originally used to confirm the absence of cytochromes in anaerobic organisms such as Streptococcus and Clostridium (9). Confirmation of the anaerobic nature of the mutant was obtained from several experiments.…”

“…which we did not detect, the main bacterial contaminants of wort and beer found by Ault (1965) and Kleyn & Hough (1971) were present in the South African lager brewery. The value of the diagnostic scheme of Ault (1965), modified by use of the benzidine test (Deibel & Evans 1960) for the recognition of lactobacilli and pediococci, the medium of Carr (1968) to distinguish between those acetic acid bacteria that oxidized acetate and those that did not, and the API 20E system for the identification of enterobacteria, was demonstrated in that with a few easily performed tests all isolates could be assigned to one of the above groups. The benzidine test was shown to be essential; without it certain catalase positive isolates of Pediococcus spp.…”
