Modular mutagenesis of exons 1, 2, and 8 of a glutathione S-transferase from the Mu class. Mechanistic and structural consequences for chimeras of isoenzyme 3-3

Abstract: Exons 1 and 2 and exon 8 of the mu class GSH transferases from rat encode sequence-variable regions 1 and 4 of mu class isoenzymes, respectively. These two of four variable regions are located at the N- and C-termini of this isoenzyme class and impinge on the active site. In order to assess the influence of these variable regions on the catalytic diversity of the class mu isoenzymes, seven chimeric isoenzymes were constructed by transplantation of the variable regions of the sequence of the type 4 subunit into… Show more

“…The K m of the high-affinity subunit of B1/B2 for GSH is one of the lowest values reported in the literature for either plant or mammalian GSTs. For mammalian GSTs an apparent K m for GSH of 27 m was reported for rat GST 4-4 with CDNB as a substrate (Zhang et al, 1992). A heterologously expressed GST from Arabidopsis exhibited an apparent K m of 80 m for GSH when assayed with CDNB (Bartling et al, 1993).…”

“…As a result, the potential carcinogens are eliminated, and DNA or other important biomolecules are protected against damage or adduct formation (Mannervik and Danielson, 1988;Zhang et al, 1992;Beckett and Hayes, 1993;Zhong et al, 1993;Ryberg et al, 1997). Glutathione S transferase-P1 (GSTP1) is a major GST, which is expressed in both normal and tumor of colon tissue (Singhal et al, 1992;Hoensch et al, 2006;Vlaykova et al, 2007).…”

“…The presence of an aliphatic side chain at this position appears to be an important steric determinant for the stereoselective behaviour of class-mu-catalyzed addition reactions with a,p-unsaturated ketones and oxiranes [118]. In rGSTM2-2, a much more efficient and stereoselective catalyst than the rGSM1-1 isoenzyme towards these substrates, Val9 is replaced with the larger Ile side chain that would extend further into the Hsite.…”
