Modulation of differentiation of rat granulosa cells in vitro by interferon-γ

Abstract: The effects of recombinant rat interferon-gamma (rRaIFN-gamma) and rat IFN (RaIFN, a mixture of IFN-gamma and -alpha) on basal and FSH-induced ovarian granulosa cell function were studied. Granulosa cells were harvested from diethylstilboestrol-treated immature rats and cultured (2 x 10(5) viable cells/well per 0.5 ml) in serum-free medium with or without treatment for 48 h. In the presence of FSH (20 ng/ml), rRaIFN-gamma (10-1000 U/ml) significantly inhibited FSH-stimulated aromatase activity (76.4 +/- 2.3% m… Show more

“…However, IFN is present in follicular fluid [24], and lymphocytes that infiltrate the ovary could provide a source of IFN [25,26]. IFN has been shown to inhibit LH receptor induction [27] and gonadotropinstimulated steroidogenesis [27][28][29], to alter inhibin production by granulosa cells [29], and to inhibit progesterone secretion by bovine luteal cells [30]. Further studies are required to determine factors relevant to the modulation of the Fas pathway in ovarian cells in vivo.…”

“…EGF, TGF-oa, FSP, interferon-y, and high doses of LH negatively regulate inhibin production in the presence of FSH and other agents that increase cAMP levels [10]. Activin and interferon-y also stimulate basal inhibin production [11][12][13]. While there is evidence for intraovarian production of most of the regulators mentioned above, it remains to be established whether or not interferon-y is made by the ovarian follicle.…”

“…This appears to be the first demonstration of an ovarian effect of -interferon. The chemically and function¬ ally unrelated gamma-interferon has been shown to decrease progesterone and increase prostaglandin secretion by bovine luteal cells (Fairchild & Pate, 1991) and to inhibit differentiation in rat granulosa cells (Gorospe et al, 1988;Xiao & Findlay, 1992).…”
