Modulation of the cannabinoid receptors by andrographolide attenuates hepatic apoptosis following bile duct ligation in rats with fibrosis

Abstract: Bile acid-induced apoptosis plays an important role in the pathogenesis of cholestatic liver disease, and its prevention is of therapeutic interest. The aim of this study was to test whether the andrographolide limits the evolution of apoptosis in a murine model of bile duct ligation (BDL)-induced hepatic fibrosis. Male Sprague-Dawley rats were divided into four groups and hepatic apoptosis was induced by BDL for 2 weeks. The BDL animals were also treated with andrographolide (50, 100, and 200 mg/kg, i.p.) dur… Show more

“…Apoptosis plays a central role in cholestatic liver injury, and this process is regulated by various players particularly the Bcl-2 family proteins [17][18][19]. Bax is a pro-apoptotic protein belonging to the Bcl-2 members that is found predominantly in the cytosol in an inactive form [17].…”

“…Caspase activities were determined using a colorimetric assay kit (BioSource International, Inc., Camarillo, CA, USA) and specific substrates (Asp-Glu-Val-Asp-pNA for caspase-3, and IleGlu-Thr-AsppNA for caspase-8) according to the previous protocol (Lee et al, 2010). The activity was expressed as a percentage relative to the control group.…”

“…It is particularly efficient at regulating immune responses and works as an anti-inflammatory agent by reducing the generation of ROS in human neutrophils (Calabrese et al, 2000;Shen et al, 2002). It has also been shown to have hepatoprotective effects in animal studies (Singha et al, 2007;Lee et al, 2010).…”

“…Andrographolide (andro) is a labdane diterpenoid, the main bioactive component of the medicinal plant Andrographis paniculata. Andro is known to have anti-fibrotic, antiinflammatory, and anti-cancerous properties (Akbarsha and Murugaian, 2000;Lee et al, 2010;Li et al, 2007;Rana and Avadhoot, 1991;Shen et al, 2009Shen et al, , 2000. Therefore, we hypothesize that andro can inhibit proliferation and migration of the lens epithelial cell line (FHL 124).…”
