Molecular allergy diagnostics refine characterization of children sensitized to dog dander

Abstract: Sensitization to an increasing number of dog allergen components and to lipocalins is associated with dog allergy. Monosensitization to Can f 5 should not be regarded primarily as a marker for dog allergy.

“…All patients reporting rhinitis (n = 40) or asthma (n = 30) upon exposure to dogs showed positive BAT responses to dog dander. No patients with a negative BAT to dog dander (n = 4) reported respiratory symptoms on exposure to dogs (rhinitis P = .007, asthma P = .048)(Table S2).Nasal provocation testing (NPT) was performed using a commercially available dog dander extract, Aquagen 100 000 SQ-U/mL (ALK-Abello) according to the protocol of Lebel et al with minor modifications 1,6. Twenty-four patients showed a positive NPT toward dog dander extract, and twenty patients had a negative NPT.…”

“…Twenty-four patients showed a positive NPT toward dog dander extract, and twenty patients had a negative NPT. The results of 14 NPTs were considered inconclusive and therefore not possible to evaluate 1,2. Positive BAT results for dog dander were found in 23 of 24 children with positive NPTs and 17 of 20 children with negative NPTs.…”

“…In the studies described here we chose to focus on one of the dog lipocalin allergens, Can f 6. Recent reports suggests that Can f 6 is a key lipocalin responsible for allergic symptoms (10).…”

“…Responses to Can f 6 are present in between 38%(11) and 56.3%(12) of dog sensitized individuals and Can f 6 allergic individuals are more likely to suffer from allergic rhinitis symptoms at a rate of 86% vs non-sensitized at 53% and those with asthma symptoms are found to be sensitized at 64% vs 38% in non-sensitized (10). Therefore, Can f 6 is a major allergen driving disease amongst those people who are allergic to dogs.…”

“…Pulmonary Medicine moderate/severe rhinitis [11]; in a Sweden population, sensitization to Can f 5 was high (61%), but it was not associated with nasal symptoms [37]. The divergent results about the clinical relevance of sensitization to dog or cat components might be explained, in part, by the lack of objective evaluations like provocation test in most of the studies.…”
