Molecular and cytological characterization of the global Musa germplasm collection provides insights into the treasure of banana diversity

Abstract: Bananas (Musa spp.) are one of the main fruit crops grown worldwide. With the annual production reaching 144 million tons, their production represents an important contribution to the economies of many countries in Asia, Africa, Latin-America and Pacific Islands. Most importantly, bananas are a staple food for millions of people living in the tropics. Unfortunately, sustainable banana production is endangered by various diseases and pests, and the breeding for resistant cultivars relies on a far too small base… Show more

“…The homogenate was filtered through a 50 μm nylon filter into a sample tube and incubated for 1–5 min on ice. Otto II buffer (1 mL) containing the fluorescent dye 4‐6‐diamidino phenyl indole (DAPI) was added before ploidy measurement using Sysmex CyFlow flow cytometer equipped with UV excitation and detectors for DAPI fluorescence (Christelová et al ., ). Molecular characterization using SSR markers targeting 19 loci was conducted as described by Christelová et al .…”

“…These results are similar to report by Samarasinghat et al (2010), where in the morphological and SSR clustering, AAB genotypes well as all other genotypes occurred in different groups. A study by Christelová et al 2016 used Molecular and Cytological systems to characterise global banana collections recommended more focus on molecular and morphological evaluation.…”

“…To date, the genomes of two banana progenitors, M. acuminata subsp. malaccensis (A genome) and M. balbisiana (B genome), and another wild banana, M. itinerans , have been sequenced with the resulting sequences revealing genome evolution peculiarities of Musaceae (D'Hont et al ., ; Davey et al ., ; Čížková et al ., ; Christelová et al ., ). This information could shed more light on Musa spp.…”
