Molecular and Physiological Detection of Cyst Nematodes on Potato During Plant Nematode Interactions

Abstract: Cyst nematodes involve over than 90 known species into six genera, G. rostochiensis and G. pallida are most important pathogens of potato crops where their economically a yield losses. The aim of present investigation is characterization the cyst nematode species (G. rostochiensis) using molecular techniques during plantnematode interaction. The molecular identification of cyst nematode was carried out using ITS PCR. The results of ITS-PCR showed two bands with different molecular sizes 238 bp and 370 bp. The … Show more

“…But there was one different band at molecular weight 49 kDa as compared with control and other sensitive cultivars at the different intervals at the same concentration of 5mM SA. Similar results were reported by lipid peroxidation detection in shoots of potato infected with cyst nematodes during intervals where; there was an increase of that enzyme after 24h to 7 days and also up to 35 days post-infection with nematodes (ELKobrosy et al, 2018). In contrast, other results mentioned that a high concentration of ethylene treatments 5mM revealed low gene expression on levels of the roots infected with cyst nematodes (ELKobrosy et al, 2022).…”

“…At 1, 24, 48, and 72 h after ethylene treatment, the roots of treated potato plants were collected to examine the defensin genes before nematode infection. After, pathogenicity assay was done for G. rostochiensis , which was isolated and identified ( Elkobrosy et al, 2018 , Elkobrosy et al, 2020 ); 60 cysts of G. rostochiensis for each pot were inoculated on potato roots. The cyst nematode was inserted into the soil near the roots.…”
