Molecular Characterization of Adult Tumors Diagnosed as Cerebellar Glioblastomas Identifies Subgroups Associated With Prognosis

Abstract: Adult tumors diagnosed as cerebellar glioblastoma (cGBM) are rare and their optimal classification remains to be determined. The aim of this study was to identify subgroups of cGBM based on targeted molecular analysis. cGBM diagnosed between 2003 and 2017 were identified from the French Brain Tumor Database and reviewed according to the WHO 2021 classification. The following molecular alterations were studied: IDH1/2, H3F3A, FGFR1, BRAF, TERT promoter mutations, EGFR amplification, MGMT promoter methylation, a… Show more

“…We processed the FBTDB and, conducted a nationwide population-based study of all histopathologically con rmed meningioma in France. Detailed methodology has previously been published [6][7][8][9][10][11][12]. In summary, FBTDB collects and records data of patients operated on for a PCNST.…”

“…Recently two diffuse glioma entities were discovered that have histone 3 (H3) alterations. Both are rare, IDH-wildtype, most often seen in children and young adults, with a prognosis similar to that of glioblastoma, IDHwildtype [18]. MGMT promotor methylation status is a biomarker of both prognostic and therapeutic significance in glioblastoma, IDH wildtype.…”

“…Although TERT promoter mutations are extremely rare in pediatric gliomas, increased TERC and TERT expression are associated with decreased ΟS in high-grade tumors [ 81 ]. Also, in cerebellar glioblastomas TERT promoter mutation and/or EGFR amplification, ATRX loss, and ALT pathway activation have been reported as independent prognostic factors with ATRX loss of expression, and ALT positivity to associated with a better outcome and TERT/EGFR alterations with a worse outcome of the patients [ 82 ].…”
