Molecular characterization of lumpy skin disease virus in Namibia, 2017

Abstract: Between January and July 2017, lumpy skin disease (LSD) outbreaks were reported in cattle in Namibia. DNA was extracted from skin biopsies taken from 32 cattle, and the RNA polymerase 30 kDa subunit (RPO30) gene of the LSD virus (LSDV) was successfully amplified by PCR. Phylogenetic analysis revealed that the newly sequenced LSDV isolates from Namibia were identical to LSDV isolates identified previously in Burkina Faso, Egypt, Greece, Niger, Serbia and South Africa. Given that only unvaccinated herds were aff… Show more

“…In an endemic environment without other organized approaches to control the disease, farmers may simply perform vaccination as a stand-alone protection measure. For many areas in sub-Saharan Africa, this has been the reality over the past decades [ 98 , 99 , 100 ], highlighting the need to adapt vaccination strategies [ 101 ]. Facing localized LSD outbreaks, countries have opted for ring vaccinations of cattle and Asian water buffalo populations surrounding the outbreak(s) [ 7 , 98 ].…”

“…The invading LSDV was similar to those discovered in China in 2019, evidenced by our phylogenetic analyses. A few viral genomic regions, GPCR gene [ 20 , 22 ], ORF011, ORF012, ORF036 [ 16 ], P32 gene [ 21 ], and RPO30 gene [ 3 , 19 ], have been used in studying phylogeny of LSDV isolates. In the present study, GPCR, RPO30, and P32 genes were employed in generating phylogenetic trees ( Figs.…”

“…Although sequencing specific regions (e.g. RPO30 and GPCR) allows to discriminate vaccine from field strains, none of these regions contain sufficient information to study possible lines of transmission within an outbreak or epidemic ( Mafirakureva et al, 2017 ; Molini et al, 2018 ; Sudhakar et al, 2020 ). Comparison of the whole genome sequences of recent LSDV isolates from the 2015–2016 epidemic in southern Europe revealed only a limited number of point mutations between the isolates ( Agianniotaki et al, 2017 ; Mathijs et al, 2020b ,).…”
