Molecular characterization of measles viruses in Turkey (2010–2011): First report of genotype D9 involved in an outbreak in 2011

Abstract: Genetic characterization of measles viruses (MVs) combined with acquisition of epidemiologic information is essential for measles surveillance programs used in determining transmission pathways. This study describes the molecular characterization of 26 MV strains (3 from 2010, 23 from 2011) obtained from urine or throat swabs harvested from patients in Turkey. MV RNA samples (n = 26) were subjected to sequence analysis of 450 nucleotides comprising the most variable C-terminal region of the nucleoprotein (N) g… Show more

“…RNA was purified from urine or throat swabs samples using a commercial viral RNAeasy Kit (Qiagen, Hilden, Germany) in accordance with manufacturer's recommendations. Like previously reported, RT‐PCR (QIAGEN) amplified 590 base pairs (bp) of the C‐terminal region of the N gene of viruses using the primer sequences n1 and n2 as described in the previous studies [Kühne et al, ; Kalaycioglu et al, ; Metin et al, ].…”

“…DNA was purified using Agencourt AMPure XP‐PCR Purification (Beckman Coulter Company, MA) as recommended by manufacturer's instructions. Sequencing reactions was set up by using cycle sequencing Quick Start Kit (Beckman Coulter) and conditions were followed as previously described [Kalaycioglu et al, , ]. Twenty microliters of DNA from each sample was subjected for sequencing with a CEQ 8000 Genetic Analyser (Beckman Coulter).…”

“…This technique can elucidate the spatial and temporal transmission dynamics of microbes, as has been done in studies of avian influenza virus [8], human immunodeficiency virus [9], hepatitis C virus [10], and Salmonella typhimurium [11]. Although there have been many epidemiological reports about inter-country transmission of measles [12,13,14,15], systematic analysis of the global transmission dynamics of MV has been limited.…”
