Molecular Characterization of Mungbean Yellow Mosaic Disease and Coat Protein Gene in Mungbean Varieties of Bangladesh

Abstract: Mungben yellow mosaic virus (MYMV), a member of the genus begomovirus causing major yield loss of mungbean. Development of MYMV tolerant/resistant varieties using an antiviral strategy may be one of the potential options to overcome MYMV diseases. In the present investigation, molecular identification of yellow mosaic disease and characterization of (MYMV) coat protein (CP) gene was attempted. Yellow mosaic symptomatic samples were identified through PCR using conserved region primers designed after alignment … Show more

“…However, MYMV, MYMIV, and HgYMV lead to YMD symptoms that are similar to those in mungbean; therefore, there is a need to confirm the specific virus(es) that lead to YMD development in mungbean, in order to accelerate the breeding of YMD-resistant genotypes [34]. Several DNA markers have been reported to be useful in detecting the presence of the specific virus leading to YMD development [35][36][37][38][39][40].…”

“…YMVs are mostly characterized either by complete sequencing or by sequencing of various DNA-A and DNA-B components. Molecular characterization of YMVs infecting mungbean in Bangladesh and Pakistan revealed 97 and 94% sequence similarity for the CP and NSP-genes of MYMIV, respectively ( Hussain et al, 2004 ; Islam et al, 2012 ). Similarly, sequence-based phylogenetic analysis of legume-infecting begomoviruses from Indonesia and Vietnam has identified Indonesian isolates as MYMIV strain-A, while Vietnam isolates as MYMV strain-B ( Tsai et al, 2013 ).…”

“…MYMIV occurs in Northern India, Pakistan, Nepal, Bangladesh, and Indonesia, while MYMV is confined to Thailand, Vietnam, and Peninsular region of India (Ilyas, Qazi, Mansoor, & Briddon, 2010;Islam et al, 2012;Shahid et al, 2012;Tsai et al, 2013). HgYMV occurs only in South India (Borah & Dasgupta, 2012;Varma & Malathi, 2003).…”

“…Three begomoviruses, MYMV, MYMIV, and HgYMV, were shown to cause mungbean yellow mosaic disease (MYMD) symptoms (Islam, Sony, & Borna, 2012;Malathi & John, 2008). MYMIV occurs in Northern India, Pakistan, Nepal, Bangladesh, and Indonesia, while MYMV is confined to Thailand, Vietnam, and Peninsular region of India (Ilyas, Qazi, Mansoor, & Briddon, 2010;Islam et al, 2012;Shahid et al, 2012;Tsai et al, 2013).…”
