MOLECULAR CHARACTERIZATION OF SOLUBLE AND MEMBRANE‐BOUND TREHALASES IN THE COTTON MIRID BUG, Apolygus lucorum

Abstract: Trehalose, a major hemolymph sugar in insects, is hydrolyzed by trehalase. We identified a soluble and a membrane-bound form of trehalase and isolated the corresponding mRNA, ALTre-1, and ALTre-2 in the cotton mirid bug, Apolygus lucorum. The deduced amino acid sequences of ALTre-1 and ALTre-2 revealed mature proteins with 643 and 617 amino acids, respectively. ALTre-1 and ALTre-2 contained trehalase signature motifs, and ALTre-2 contained a putative transmembrane domain near the C-terminus, suggesting that AL… Show more

“…The trehalase content in the different organs of H. armigera was measured, and the enzyme was found primarily in the midgut, which was consistent with that in other insects (Tan et al, ; Xie, Yang, Dou, & Wang, ). According to the alternation of the three carbohydrates and the related enzymes, all of the three β‐glucosides can inhibit trehalase, and it can also be proven that santonin was the most effective inhibitor in H. armigera .…”

“…In insects, trehalase is one of the most important enzymes of carbohydrate metabolism, which is the central part of these organisms’ metabolism. Trehalase is classified into two types: soluble and membrane trehalase (Tan et al, ). This enzyme fuels energy‐demanding synthetic reactions (Beitner & Rivka, ) and provides necessary building blocks for various anabolic pathways (Nakano & Sacktor, ).…”

“…The relative gene expression levels of AlEcR-B were represented by relative quantification values calculated by the realtime quantitative PCR and the 2 2DDCt method (Livak & Schmittgen, 2001). The mRNA expression profiles of AlTre-1 and AlTre-2 across developmental stages (first-fifth-instar nymph and female adult) were assayed using the method of Tan et al (2014b).…”
