Molecular cytogenetic characterization of two murine cancer cell lines derived from salivary gland

Abstract: Here two murine salivary gland cancer (SGC) cell lines WR21 and SCA-9 were studied for the first time in detail by high-resolution molecular cytogenetic approaches. This study revealed that these cell lines are models for human SGCs of initial stage myoepithelioid or mucoepidermoid (WR21) and of advanced stage mucoepidermoid (SCA-9) tumors. Besides, three genes most likely playing a role in SGC development (FGF10, ELAVL1/HUR and SEL1) were identified. All of them were involved in translocation events in these … Show more

“…Thus, in this study, the three frequently used murine BC cell lines C-127I, EMT6/P and TA3 Hauschka were for the first time characterized on the molecular cytogenomic level. The feasibility of the applied scheme was shown in several previous studies [ 20 , 36 , 37 , 38 , 39 ].…”

“…mFISH using whole chromosome painting probes revealed the general characteristics of the cell lines, like the ploidy, clonal and nonclonal changes as well as numerical and intrachromosomal structural aberrations. By mcb interchromosomal alterations as duplications, deletions or inversions, and chromosomal breakpoints involved could be uncovered, as previously reported [23][24][25][26][27][28]. The aCGH data was aligned with the FISH results and also used to determine breakpoints of unbalanced rearrangements (Additional file 1: Table 1).…”

“…Accordingly, especially murine tumor cell lines are favorite model systems to perform basic research studies, as well as such towards testing of new medications [ 8 , 33 ]. Our group characterized already several murine tumor cell lines by the same test strategy as applied here, and thus delivered previously not available but urgently necessary genetic basic data, including chromosome numbers, observable breakpoints and copy number changes [ 1 , 16 , 17 , 19 , 34 , 35 ]. Here a commercially available murine BC-cell line was characterized for the first time, cell line JC, which was yet rarely used in research.…”

“…JC cell line presents a near-tetraploidy karyotype, which is typically observable, especially in human [ 36 ], however, only present in about 50% of murine tumor cell lines [ 1 , 16 , 17 , 19 , 34 , 35 ]. Polyploidy promotes malignant transformation of mammary cells and increases cell resistance to drugs [ 36 ]; still, it is also a known adaptation of cells to cell culture conditions.…”
