Molecular detection of Trypanosoma sp. and Blastocrithidia sp. (Trypanosomatidae) in phlebotomine sand flies (Psychodidae) in the Federal District of Brazil

Abstract: Introduction:This study describes the occurrence of trypanosomatids in phlebotomines in Brasília, Brazil. Methods: Two hundred and ten females of 13 sand fl y species were analyzed by polymerase chain reaction (PCR) using different molecular markers (D7 24Sα rRNA, kDNA, and ITS1) and sequencing. Results: PCR revealed trypanosomatid-positive samples from Nyssomyia whitmani and Evandromyia evandroi, which were negative by kDNA and ITS1 Leishmania-specifi c PCRs. DNA sequence analysis of D7 24Sα rRNA amplicons in… Show more

“…In particular, Trypanosoma platydactyli was described in S. minuta [42], and the infection by a Trypanosoma belonging to Trypanosoma theileri group with very high homology to other trypanosomes detected in European cervids was recently reported in Phlebotomus perfiliewi [43]. In the Mediterranean area, Trypanosoma nabiasi, a rabbit trypanosome, and its co-infection with L. infantum was found in P. perniciosus female sand flies caught in the context of human leishmaniosis outbreak in Madrid [44], and natural infection of sand flies by trypanosomes of lizards, amphibians, birds and rodents has been already reported mainly from the American continent and Asia [45][46][47][48]. Despite Trypanosoma spp.…”

“…We expected to capture sandflies in this area because they are frequent in gallery forests of the FD (Ferreira, Macedo, et al, ; Rapello et al, ). The absence of Leishmania ‐infected sandflies may be directly related to the low number of specimens captured and the low rate of infection by trypanosomatids in the studied area (Ferreira, Minuzzi‐Souza, et al, ).…”

“…cruzi in the forest adjacent to the Brasília Zoo (Gurgel‐Gonçalves et al, ). Leishmania infantum ‐infected dogs have also been confirmed in FD (Cardoso et al, ), where sandflies are frequent in gallery forests (Ferreira, Minuzzi‐Souza, et al, ; Rapello et al, ) and houses (Carvalho, Bredt, Meneghin, & Oliveira, ). Studies of trypanosomatid‐infected mammals and vectors (triatomines and sandflies) in zoos are important for the establishment of surveillance and control measures for leishmaniasis and Chagas disease, including valuable information for animal health and management strategies such as translocations.…”

“…Ecological host-fitting, involving opportunistic host switching mediated by vectors' feeding patterns within an ecological niche, is thought to be a prevailing mode of trypanosome evolution [ 65 ]. Reptilian trypanosomes are transmitted by sand fly vectors [ 65 66 ], with reports from Amazonia ( Viannamyia tuberculate [ 67 ]) as well as central Brazil ( Evandromyia evandroi [ 68 ]). Shared microhabitat use among bats, reptiles and sand flies potentiates spill-over of the parasite.…”
