Molecular Mechanism of Brassinosteroid Perception by the Plant Growth Receptor BRI1

Abstract: Brassinosteroids (BRs) are essential phytohormones which bind to the plant receptor, BRI1, to regulate various physiological processes. The molecular mechanism of the perception of BRs by the ectodomain of BRI1 remains not fully understood. It also remains elusive why a substantial difference in biological activity exists between the BRs. In this work, we study the binding mechanisms of the two most bioactive BRs, brassinolide (BLD) and castasterone (CAT) using molecular dynamics simulations. We report free en… Show more

“…This coupling of binding and hydrolysis makes it difficult to elucidate the effects of substrate binding on signaling independently of subsequent steps. A powerful method that can be used to characterize the strigolactone binding process is molecular dynamics (MD) simulations ( 20 , 21 , 22 , 23 , 24 , 25 , 26 ). When used with Markov state models (MSMs), simulations can provide detailed kinetic and thermodynamic information about ligand-binding processes at atomic-level resolution ( 27 , 28 , 29 , 30 , 31 , 32 ).…”

“…Finally, recent work from our group has tried to provide a comprehensive structural and dynamic view of plant hormone signaling for ABA 33 , Brassinosteroid 40,56,57 and Strigolactone 58 hormone receptors in plants. These physical insights into the structural, dynamic and energetic basis of the activation of hormone receptors in Arabidopsis thaliana, which are challenging to obtain with experimental approaches.…”

“…This study used 24-epicastasterone (ECS), which is a precursor of castasterone, an end product of BR biosynthesis pathway in rice [ 35 ], and has been shown to be biologically active but less so than brassinolide [ 36 ]. ECS (Yuanye Biology, Shanghai, China), was dissolved in 80% ethanol.…”
