Prediction of Pan-Specific B-Cell Epitopes From Nucleocapsid Protein of Hantaviruses Causing Hantavirus Cardiopulmonary Syndrome

Hantaviruses are emerging viral pathogens that causes hantavirus cardiopulmonary syndrome (HCPS) in the Americas, a severe, sometimes fatal, respiratory disease in humans with a case fatality rate of ≥50%. IgM and IgG-based serological detection methods are the most common approaches used for laboratory diagnosis of hantaviruses. Such emerging viral pathogens emphasizes the need for improved rapid diagnostic devices and vaccines incorporating pan-specific epitopes of genotypes. We predicted linear B-cell epitopes for hantaviruses that are specific to genotypes causing HCPS in humans using in silico prediction servers. We modeled the Andes and Sin Nombre hantavirus nucleocapsid protein to locate the identified epitopes. Based on the mean percent prediction probability score, epitope IMASKSVGS/TAEEKLKKKSAF was identified as the best candidate B-cell epitope specific for hantaviruses causing HCPS. Promiscuous epitopes were identified in the C-terminal of the protein. Our study for the first time has reported pan-specific B-cell epitopes for developing immunoassays in the detection of antibodies to hantaviruses causing HCPS. Identification of epitopes with pan-specific recognition of all genotypes causing HCPS could be valuable for the development of immunodiagnositic tools toward pan-detection of hantavirus antibodies in ELISA. 22 23 24 25 2 26 Abstract 27 Background: ANDV, agent of hantavirus cardiopulmonary syndrome, enters through 28 integrin cell protein. A change from leucine-to-proline at residues 33 in the PSI-domain 29 (L33P) inhibits ANDV recognition. We assessed the association between this human-30 variant and ANDV infection.31 Results: We defined susceptible genotype to "TT" (coding leucine) and protective "CC" 32 (coding proline). TT was in 89.2% (66/74) of a first cohort of ANDV-cases and in 60% 33 (63/105) of exposed close-household contacts who remained unifected (p<0.05).34 Protective genotype was absent in all 85 ANDV cases in both cohorts and was present at 35 11.4% in exposed close-household contacts who remained uninfected. Logistic regression 36 modeling to become a case had an OR 6.2-12.6 (p<0.05) in presence of TT and ANDV 37 well-known risk activities. Andes orthohantavirus (ANDV), is the etiological agent of HCPS in Chile 56 and South of Argentina, and is the only hantavirus recognized to be transmited person-57 to-person. Here we try to understand the role of one human genetic variant of 3-integrin 59 in susceptibility to ANDV-infection. We compared, exposed infected and non infected 60 subjects, and their genotype differences regarding a 3 integrin variant that change a 61 Leucine to Proline at PSI domain of the receptor. Leucine and proline aminoacids at the 62 PSI domain, turn the cell susceptible or resistant to ANDV-infection, respectively. Hantaviruses are the causative agents of hantavirus pulmonary syndrome in humans in the Americas; The primary reservoirs are in the rodents of the subfamily Sigmodontinae. In South America, cases of hantavirus pulmonary syndrome caused by numerous viral genotypes have been diagnosed. In Colombia, different serological studies have reported the circulation of hantavirus in humans and rodents. These viruses act in an intimate association with a rodent species that serves as a reservoir and have a distribution around the wild rodent, being limited to a specific geographic region. In South America, the first HPS-associated hantavirus was described in 1993 in Brazil and was called Juquitiva and from 1993 to 2012, more than 1400 cases had been identified in Brazil. This syndrome should be suspected in all patients with respiratory distress syndrome of unclear etiology, in areas endemic for the disease, especially if accompanied by fever, marked leukocytosis and thrombocytopenia and bilateral interstitial infiltrates. Hemorrhagic febrile syndrome has not yet been described in the Americas. There are no clinical or laboratory signs that are pathognomonic of hantavirus infection. The treatment is based on adequate hydration, use of antipyretics and anti-inflammatories and patients with signs of severity should establish a more aggressive management. Triage is indispensable, patients with co-morbidities have a higher mortality risk and therefore should be hospitalized. Future research in Colombia should be directed to multidisciplinary studies that include viral isolation, different clinical forms of case presentation, epidemiological differences, risk factors, and taxonomy of viruses and rodents.Keyswords: Arenavirus, bunyaviridae, reservoirs, rodentia, public health (Source: DeCS). En Suramérica, se han diagnosticado casos del síndrome pulmonar por hantavirus y se han identifico numerosos genotipos virales. En Colombia, se han publicado diferentes estudios serológicos que reportaron la circulación de hantavirus en humanos y roedores. Estos virus actúan en una íntima asociación con una especie de roedor que le sirve de reservorio y presentan una distribución alrededor del roedor silvestre, limitándose a una región geográfica especifica. En Suramérica, el primer hantavirus asociado a SPH fue descrito en 1993 en Brasil y se denominó Juquitiva y desde 1993 hasta el 2012, se habían identificado más de 1400 casos en Brasil.
