“…The premolt stage comprises the preparatory events required for ecdysis, such as an increase in circulating ecdysteroids, separation of the old cuticle from the underlying hypodermis (apolysis), and, in some crustacean species, the accumulation of calcium in the gastroliths, paired disk-like calcium-storage organs enveloped by epithelial cells, which are situated in the cardiac stomach wall (McWhinnie, 1962;Travis, 1960;Ueno, 1980). The surge in circulating ecdysteroids triggers a programmed sequence of expression of different genes and synthesis of proteins in a variety of tissues, including the hypodermis and the hepatopancreas (Stringfellow and Skinner, 1988;Traub et al, 1987). This increase in circulating ecdysteroids is followed by a decrease in the levels of circulating ecdysteroids just before ecdysis, a step that is essential for the shedding of the cuticle; with ecdysis being delayed in the absence of theses changes in the ecdysteroids levels (Chang, 1993).…”

“…We show electrophoretic patterns of proteins and compare the proteins present in anecdysial, proecdysial, and metecdysial exoskeletons and exuviae of G. lateralis. The distribution of a number of the proteins in one or more layers of the exoskeleton (data shown here), as well as the patterns of their synthesis (Stringfellow and Skinner, 1988;Paulson and Skinner, 1991 ) and degradation Skinner, 1987, 1988), make them favorable candidates for the investigation of genes specifically activated during the molting process.…”

“…Gel electrophoresis A discontinuous gel system was used for ID gels (Laemmli, 1970). Separating gels were gradients of 5-15% polyacrylamide (Stringfellow and Skinner, 1988) or 9-18% polyacrylamide, the latter contained 0-10% glycerol (O'Brien and Skinner, 1988); stacking gels were 4.5%. acrylamide.…”

“…Ecdysteroids, the moulting hormones common to all arthropods, are synthesized in the crustacean Y-organs, endocrine steroidogenic glands of ectodermal origin (reviewed in Lachaise et al, 1993). The increase in ecdysteroid synthesis at the onset of premoult leads to a surge of circulating 20-hydroxyecdysone (20E) that triggers a programmed synthesis of proteins in the epidermis and the hepatopancreas (Bielefeld et al, 1986;Traub et al, 1987;Stringfellow & Skinner, 1988). For successful ecdysis, a decrease in the 20E level is required immediately prior to moulting; without this drop ecdysis is delayed (Chang et al, 1993).…”
