Monoclonal antibodies that detect live salmonellae

Abstract: Nine immunoglobulin G and nine immunoglobulin M murine monoclonal antibody-producing hybridomas reactive with live SalmoneUla bacteria were obtained from several fusions of immune spleen cells and Sp2/0 myeloma cells. The antibodies were selected by the magnetic immunoluminescence assay. The monoclonal antibodies were reactive with serogroups A, B, Cl, C2, D, E, and K and SalmoneUla choleraesuis subsp. diarizonae. Each monoclonal antibody proved to be reactive with a distinct serotype. Clinical isolates belong… Show more

“…Monoclonal antibodies. The generation of serogroup-specific monoclonal antibodies has been described by Torensma et al (16). The following six monoclonal antibodies were used in this study: monoclonal antibody 47.15 (IgM), specific for serogroup A; monoclonal antibody 71.40 (IgGl), specific for serogroup B; monoclonal antibody 72.4 (IgG3), specific for serogroup Cl; monoclonal antibody 35.71 (IgGl), specific for serogroup C2; monoclonal antibody 41.11 (IgGl), specific for serogroup D; and monoclonal antibody 42.77 (IgG3), specific for serogroup E.…”

“…in which monoclonal antibodies that are specific for live Salmonella spp. and a PCR based on Salmonella-specific primers chosen in the origin of bacterial chromosome DNA replication (oriC) were combined (16,19). The use of the MIPA allows the circumvention of Taq polymerase inhibition by components of food, enrichment media, or large amounts of non-Salmonella DNA.…”

“…Lower doses or shorter schedules appeared to be ineffective for raising antibodies to live bacteria. At 3 days after the intravenous booster injection the spleen cells were collected and fused with the Sp2/0 myeloma cell line as described previously (21,22). Part of the fused cell mixture (fusion 55) was cultured for 5 days.…”

“…booster of a mixture of three clinical isolates of the desired serotype containing 10 8 gentamicin-treated bacteria was given. Spleen cells were collected 3 days after the booster and fused with the SP2/0 myeloma cell line, using electrofusion (31). A fusion pulse of either 2 or 3 kV/cm was given with a duration of 10 s. Hybridoma supernatant fluids were assayed by agglutination to detect the presence of specific antibodies against either E. coli O18KϪ or E. coli K5.…”
