Centaurium erythraea is medicinal plant with high content of secoiridoid glycosides and xanthones with gastroprotective, hepatoprotective and antioxidative properties. In vitro culture based C. erythraea production would offer sustainable and economically feasible alternative to wild plant harvest provided that comparable productivity could be achieved in artificially propagated plants. To identify most productive C. erythraea system the secoiridoid glycoside and xanthone production and profiles using liquid chromatography-mass spectrometry were assessed in in vitro shoot, callus and cell suspension culture-derived material in comparison with wild C. erythraea plants.In vitro shoot culture was the most productive system for C. erythraea secoiridoid production (total yield 86 mg g -1 dw) and cell culture was the most productive system for phenolic compound production (19 mg GAE g -1 dw). The effect of elicitors yeast extract, methyljasmonate (MeJA) and chitosan on secondary metabolite yield indicated that elicitation with MeJA and chitosan significantly decreased yield of sweroside and eustomin, respectively. Significant quantitative differences of major secoiridoids and xanthones indicated that further methodological improvements for increasing yield of major secondary metabolites of in vitro origin need to be explored for in vitro cultivation to become a feasible alternative to C. erythraea wild plant harvest.

Since the 1980s, there has been a significant emphasis in biotechnology on the utilisation of medicinal plants as a source of raw materials for the pharmaceutical industry. Moreover, medicinal plants have been identified as a potential alternative source of essential compounds with a wide range of applications, including those involved in producing medications, nutraceuticals, food additives, cosmeceuticals, natural pigments, and preservatives. The plant micropropagation system is an ideal solution to the problems caused by conventional exploitation since it may simultaneously provide uniform biomass as a source of bioactive secondary metabolites and ex situ conservation of uncommon or endangered plant species (dual strategies of micropropagation). In honour of the Polish Botany Society’s Centenary anniversary in 2022, this article summarises the development of an in vitro propagation method for selected medicinal plants by Polish researchers and botanists.

Plants are an immense source of phytochemicals with therapeutic effects and are widely used as life-saving drugs, and other products of varied applications. Plant tissue culture is a unique technique employed under aseptic conditions from different plant parts called explants (leaves, stems, roots, meristems, etc.) for in vitro regeneration and multiplication of plants and synthesis of secondary metabolites (SMs). Selection of elite germplasm, high-producing cell lines, strain enhancements, and optimization of media and plant growth regulators may lead to increased in vitro biosynthesis of SMs. Interventions in plant biotechnology, like the synthesis of natural and recombinant bioactive molecules of commercial importance, have attracted attention over the past few decades; and the rate of SMs biosynthesis has increased manifold than the supply of intact plants, leading to a quick acceleration in its production through novel plant cultures. Over the years, the production of SMs in vitro has been enhanced by standardising cultural conditions, selection of high-yielding varieties, application of transformation methods, precursor feeding, and various immobilization techniques; however, most often, SM production is the result of abiotic or biotic stresses, triggered by elicitor molecules like natural polysaccharides (pectin and chitosan) that are used to immobilize and cause permeabilization of plant cells. In vitro synthesis of SMs is especially promising in plant species with poor root systems, difficulty in harvesting, unavailability of elite quality planting material, poor seed set and germination, and difficult to propagate species. Thus, the present article reviews various biotechnological interventions to enhance commercially precious SMs production in vitro.
