Mouse Germ Cell Developmentin-vivoandin-vitro

Abstract: In mammalian development, primordial germ cells (PGCs) represent the initial population of cells that are committed to the germ cell lineage. PGCs segregate early in development, triggered by signals from the extra-embryonic ectoderm. They are distinguished from surrounding cells by their unique gene expression patterns. Some of the more common genes used to identify them are Blimp1, Oct3/4, Fragilis, Stella, c-Kit, Mvh, Dazl and Gcna1. These genes are involved in regulating their migration and differentiation… Show more

“…POU5F1 (previously known as octamer-binding transcription factor 3/4, OCT4) was originally identified as an ES cell and a germ line-specific marker (Okamota et al 1990, Scholer et al 1990). The expression of Pou5f1 is regulated by a proximal promoter, a germ-specific distal enhancer, and a retinoic acidresponsive element (Saiti & Lacham-Kaplan 2007). At gastrulation, Pou5f1 expression is down-regulated and thereafter is maintained only in primordial germ cells (Yeom et al 1996).…”

“…Table III summarizes the duration of murine testicular germ cell development and the type of genes expressed during embryogenesis and shows that PGCs, with their specific genes, first appear on the 7th -8th embryonic day; however their differentiation to germ stem cells occurs between the 13th and the 17th embryonic day (Saiti and Lacham-Kaplan 2007).…”

“…Clásicamente las CGP en el embrión son marcadas con TNAP, sin embargo este marcador no es específico para estas células ya que también es expresado por las cé-lulas somáticas que las rodean (Saiti & Lacham-Kaplan, 2007). Su expresión en las CGP de ratón se puede observar desde los 7,25 dpc.…”
