Mulberry Fruit Extract Ameliorates AdipogenesisviaIncreasing AMPK Activity and Downregulating MicroRNA-21/143 in 3T3-L1 Adipocytes

Abstract: Mulberry (Morus alba L.) fruits have long been used in traditional medicine and as edible berries in many countries. This study investigated the antiadipogenic effect of high hydrostatic pressure mulberry fruit extract (MFE) during 3T3-L1 adipocyte differentiation. MFE decreased lipid and triglyceride accumulation and glycerol-3-phosphate dehydrogenase activity. The mRNA expression levels of genes related to adipogenesis, such as the adipocyte protein 2, proliferator-activated receptor-c, and CCAAT/enhancer bi… Show more

“…In contrast, dMF had little impact on TC and LDL. These observations generally reflect mouse studies using statins and mulberry extracts that reduce obesity, 28 , 29 , 30 , 31 , 32 , 33 diabetes 34 , 35 , 36 , 37 , 38 and hyperlipidemia 21 , 30 , 39 …”

“…In humans, MIR21 gene is located in the q23.1 region of chromosome 17, immediately downstream of the vacuole membrane protein-1 (VMP1) gene and resides within exon 10 of the VMP1 gene (Ribas et al, 2012). Various transcription factors may be involved in the regulation of miR-21 expression, including activator protein 1 (AP-1), Ets/PU.1, CCAAT/enhancer-binding protein α (C/EBPα), nuclear factor I (NF-I), serum response factor (SRF), p53, and STAT3 (Misawa et al, 2010;Luo et al, 2014;McClure et al, 2014;Collins and Hess, 2016;Lee and Kim, 2020). However, different transcription factors have different regulatory signaling pathways for miR-21 expression.…”

“…These effects were achieved by regulating the expression of mRNA involved in de novo cholesterol biosynthesis, cholesterol efflux, bile acid synthesis and HDL-C formation [110]. In 3T3-L1 cells, incubation with mulberry fruit extract could activate AMPK and inhibit adipogenesis genes, leading to the decrease of intercellular lipid accumulation and TG content [111].…”
