Multiparametric Flow Cytometry of Human Squamous Cell Carcinoma Lines from the Head and Neck

Abstract: Squamous cell carcinomas of the head and neck consist of heterogeneous cell populations. The purpose of the present study was to investigate whether established cell lines from human head and neck cancers under chemotherapy behaved similarly to tumors in patients during in vivo treatment. This is of interest in terms of improvements of chemotherapeutic protocols and understanding of the mechanisms of cytotoxic drug resistance. Permanent squamous carcinoma cell lines of the larynx (HLaC 78, 79), parotid gland (… Show more

“…However, in 2-dimensional (2D) monolayer cultures cells tend to loose tissue-specific properties (2). 2D culture models therefore seem unsuitable to mimic the behaviour of cells in a natural, 3-dimensional (3D) microenvironment, particularly for testing of growth or invasion inhibiting substances, just as proposed in 1988 (3). In many aspects spheroids resemble the solid, nonvascularized tumour tissue and spheroidal cells show higher morphological and functional differentiation than monolayer cells.…”

“…In the mid-eighties Valet began to develop a classification system (CLASSIF1) for the individual recognition of disease progression (26) (27,28,29,30). Around this time he began to consolidate the results of his findings, which ultimately merged into the concept known as ''Predictive Medicine by Cytomics'' (31).…”

“…In the mid-eighties Valet began to develop a classification system (CLASSIF1) for the individual recognition of disease progression (26) using the numerous quantitative data acquired by flow and imaging instrumentation. He demonstrated the importance of the multiparametric approach in combination with cytometry in a multitude of clinical and biological examples (27,28,29,30). Around this time he began to consolidate the results of his findings, which ultimately merged into the concept known as ''Predictive Medicine by Cytomics'' (31).…”
