Many Arctic biomes, which are populated with abundant and diverse microbial life, are under threat: climate change and warming temperatures have raised concerns about diversity loss and possible emergence of pathogenic microorganisms. At present, there is little information on the occurrence of Arctic virulence-associated phenotypes. In this study we worked with 118 strains of bacteria (from 10 sampling sites in the Arctic region, located in Greenland and the Svalbard Archipelago) isolated using R2A medium. These strains belong to 4 phyla and represent 36 different bacterial genera. Phenotypic resistance to 8 clinically important antimicrobials (ampicillin, chloramphenicol, ciprofloxacin, cefotaxime, erythromycin, imipenem, kanamycin, and tetracycline) and thermotolerance range were determined. In addition, a screening of all isolates on blood agar media and erythrocytes suspension of bovine and sheep erythrocytes for virulence-linked hemolytic activity was performed. Although antimicrobial resistance profiles varied among the isolates, they were consistent within bacterial families and genera. Interestingly, a high number of isolates (83/104) were resistant to the tested concentration of imipenem (4 mg/L). In addition, one third of the isolates showed hemolytic activity on blood agar, however, in only 5% of the isolates hemolytic activity was also observed in the cell extracts when added to erythrocyte suspensions for 60 min. The observed microbial phenotypes contribute to our understanding of the presence of virulence-associated factors in the Arctic environments, while highlighting the potential risks associated with changes in the polar areas in the light of climate change.

Surplus sludge produced by sewage treatment plants is one of the important repositories of antibiotic. In order to reduce the adverse effect of the antibiotics on the environment, the sludge drying reed bed was used to treat the sludge. The experiment investigated the removal efficiency of antibiotics in the process of sludge stabilization by the dry bed with ventilation structure and the sludge drying reed bed with and without ventilation structure. The research project spent four years, including three years loading period and one year natural stabilization period. In this paper, the degradation of target antibiotics (roxithromycin, azithromycin and oxytetracycline) during the natural sludge stabilization period was studied, the effects of different conditions on the removal efficiency of antibiotic were analyzed. The results show that the sludge drying reed bed can effectively remove roxithromycin, azithromycin and oxytetracycline in the sludge, the reed and ventilation structure plays an obvious role for promoting antibiotics removal in the system.

Mercury tolerance and biosorption in bacteria isolated from Ny‐Ålesund, Svalbard, Arctic

Mercury tolerant bacteria Pseudarthrobacter oxydans strain MM20 and Pseudomonas frederiksbergensis strain SS18 were isolated from the tundra ecosystem of Ny-Ålesund, Svalbard, where commercial exploitation of the coal existed till 1960s. Minimum inhibitory concentration (MIC), mercury removal, mercury biosorption, and antibiotic resistance of these strains were analyzed. P. frederiksbergensis strain SS18 showed high tolerance (2.0 ppm) to mercury than P. oxydans strain MM20 (1.5 ppm). Mercury removal and biosorption studies were carried out in liquid media containing 1.0 ppm mercury. More than 90% of mercury was removed from the culture media by the selected strains. The mercury biosorption assay revealed that a part of mercury was accumulated in cell pellets and was 22 and 25% respectively for P. oxydans strain MM20 and P. frederiksbergensis strain SS18. Fourier transform infrared study revealed that alkyl halide, alkynes, alcoholic, aliphatic and aromatic amines, alkanes, nitro compound, primary amines, carboxylic acid, alkenes, and amide groups play a major role in the development of tolerance towards mercury. Out of eleven antibiotics tested, P. oxydans strain MM20 was found to be resistant to lincomycin and novobiocin while P. frederiksbergensis strain SS18 was found to be resistant to seven antibiotics. Our study demonstrates that under experimental conditions, bacterial isolates undergo detailed structural and functional changes to tolerate as well as immobilize toxic elements like mercury.
