Multiple Parameter Optimization for Maximization of Pectinase Production by Rhizopus sp. C4 under Solid State Fermentation

Abstract: Abstract:A novel strain Rhizopus sp. C4 was isolated from compost for the production of pectinase. Cultivation of Rhizopus sp. C4 on orange peel substrate under various solid-state fermentation (SSF) conditions was evaluated for pectinase yield along with the enzyme activity profile as a potential, low-cost alternative to submerged-liquid fermentation. Response surface methodology (RSM) was employed to optimize various environmental parameters for pectinase production. Various parameters, namely temperature, m… Show more

“…Wheat bran has been reported to be mainly used by many researchers as the best inducer for xylanase and pectinase production . Apart from wheat bran, various other agroresidues such as rice bran , barley bran , corncob , wheat straw , rice straw , sorghum straw , maize straw , paddy husk , cassava bagasse , apple pomace , banana waste , sugarcane baggases , sweet sorghum baggase , and oil‐palm‐empty‐bunches have been used for xylanase production and orange/citrus peel , rice straw, orange pulp , banana peel , sugarcane bagasse , sugar beet pulp , and pineapple residue for pectinase production by different microorganisms.…”

“…The absorbance of the filtered juices was recorded (650 nm ) to elucidate the effect of pectinase on fruit juices. Enzyme was not applied in the control and commercial pectinase (Sigma, U.S.A) was used as standard …”

“…Likewise, the achieved results are in agreement with those stated by Tari et al(2007) where they indicated that the pvalue reveals the correlation amongst the variables, and the response variable, pvalue lower than 0.05, shows that the practical model is significant. The model Fvalue and Probability value (P model > F) of 170.45 and <0.0001 respectively, for pectinase activity, show that the applied model is highly significant(Handa et al (2016). Likewise,(Asia et al 2019) reported that the significance of the model (p<0.05) was determined by Analysis of variance (ANOVA) that indicated an F-value of 8.22.…”
