Multiple Signaling Pathways Regulate Yeast Cell Death during the Response to Mating Pheromones

Abstract: Mating pheromones promote cellular differentiation and fusion of yeast cells with those of the opposite mating type. In the absence of a suitable partner, high concentrations of mating pheromones induced rapid cell death in ϳ25% of the population of clonal cultures independent of cell age. Rapid cell death required Fig1, a transmembrane protein homologous to PMP-22/EMP/MP20/Claudin proteins, but did not require its Ca 2؉ influx activity. Rapid cell death also required cell wall degradation, which was inhibited… Show more

“…Different markers can be used in yeast cells to distinguish between non-apoptotic and apoptotic death. In the case of non-apoptotic death, the process is accompanied by an increase in plasma membrane disorganization and permeability to staining agents such as propidium iodide and methylene blue (Dudgeon et al, 2008;Zhang et al, 2006). In experiments using the latter agent, plasma membrane-compromised cells began appearing at early times of treatment and progressively increased in number (Fig.…”

“…12,36 In contrast, death caused by elevated concentrations of pheromone lacks certain hallmarks of apoptosis. 42 Of note, sporulation following meiosis of diploid cells is coupled to apoptosis as well. 14 Interestingly, both apoptosis and meiosis in yeast have been demonstrated to involve H2B phosphorylation.…”

“…Also during sporulation yeast cells have been shown to undergo apoptotic cell death when respiration is enhanced through pre-culturing the cells in glycerol media [35]. To the contrary, Zhang et al proposed a rather necrotic type of death upon pheromone treatment of yeast cells [36].…”
