Multiplex flow-through immunoassay formats for screening of mycotoxins in a variety of food matrices

Abstract: Two multi-analyte flow-through immunoassay formats for rapid detection of mycotoxins in a variety of food matrices (peanut cake, maize and cassava flour) were developed and evaluated. The selected food matrices are typical staple foods and export products for most low-income communities around the world. The assay formats included a gel-based and a membrane-based flow-through assays and were based on the principle of indirect ELISA. Using the same immunoreagents, the performance characteristic of both assays w… Show more

“…Analytical methods are generally well established for many mycotoxins; currently liquid chromatography (LC) based methods coupled to tandem mass spectrometry (MS) are probably the most widely applied and preferred techniques (as also shown from the analyzed datasets below). The current trend in mycotoxin analysis seems to be on the one hand to reach ever lower concentration levels and multi-mycotoxin and full metabolite profiles (Arroyo-Manzanares et al 2018;Brouwer-Brolsma et al 2017;de Lourdes Mendes de Souza et al 2013;Doppler et al 2016;Adekoya et al 2019;Malachov a et al 2014Malachov a et al , 2018Njumbe Ediage et al 2015;Monbaliu et al 2010;Rofiat et al 2015;Shephard et al 2013;Simader et al 2015;Uhlig et al 2013;Varga et al 2013;Warth et al 2012;, and on the other hand to determine the compliance at the statutory levels by fast on-site techniques (Dorokhin et al 2011;EC 2006b;Ediage et al 2012;Joshi et al 2016;Peters et al 2017;Sieger et al 2017). The current multi-analyte and multi-class analytical methods are capable of a high throughput of samples making larger amounts of results available than ever before in a short period of time.…”

“…The use of multiplex immunochemical techniques, such as ELISAs (Adrian et al 2008), lateral flow dipsticks (Kolosova et al 2007), immunochips , microarrays (Hu et al 2013), electrochemical immunosensors (Kuang et al 2010) membrane- (Njumbe Ediage et al 2012) and gel-based immunoassays (Beloglazova et al 2010a), is currently increasing and becoming increasingly popular. However, most of the described multiplex immunoassays were developed by placing immunoreagents specific toward different analytes on separate spots (or test-zones, tubes or wells) within one test system.…”

“…Many conventional analytical methods are very sensitive and accurate, yet technically complex, time-consuming, require costly and sophisticated instrumentation, and do not allow for field portability or high-throughput analysis [5][6][7]. The lateral flow immunoassay is a user-friendly format in terms of simplicity and cost-effectiveness and allows for rapid on-site testing [8].…”
