Multiplex‐PCR for the detection of viruses in the CSF of infants and young children

Abstract: We aimed to establish the utility of the Fast-track diagnostics Viral meningitis multiplex PCR kit for the diagnosis of central nervous system infection in infants. The multiplex assay had reduced sensitivity for the detection of enterovirus, the predominant pathogen in young infants, when compared to our in-house singleplex PCR. In our infant population, multiple singleplex PCR assays perform better than a multiplex assay for the detection of CSF viruses. J. Med. Virol. 89:559-561, 2017. © 2016 Wiley Periodic… Show more

“…This might raise the question if using in-house PCR assays for HSV, enterovirus and HHV-6 instead of using the mPCR, may be more cost effective in some institutions. Some authors even implied that the sensitivity of a singleplex assay to detect viral agents is greater than that of a mPCR [15,17,44]. However, this approach involves the risk of missing a viral pathogen and requires a certain level of medical experience.…”

“…Multiplex assays for diagnosis of meningitis are commercially available, for example, BioFire, SeeGene and Fast Track diagnostics, and Clart Entherpex Genomica . Panels used in these assays include HSV1/HSV2 but not MTB.…”

“…The prevalence of MTB observed in our study (1.5%) is close to a report from Niger where 1.9% of confirmed cases of bacterial meningitis were caused by MTB. 50 Multiplex assays for diagnosis of meningitis are commercially available, for example, BioFire, 51,52 SeeGene 53 and Fast Track diagnostics, 54 and Clart Entherpex Genomica. 30 DNA.…”

“…Reversetranscription PCR (RT-PCR) to detect the viral nucleic acid in the CSF of PVM patients revealed a 100% sensitivity rate and proved far superior to the virus isolation technique in both EVsand HPeV-positives cases of PVM (23,24). Multiplex PCR is now frequently used in clinical and epidemiological investigations to identify the causative virus in PVM patients (162,(186)(187)(188). Additionally, approval of some molecular diagnostic kits, such as FilmArray R BioFire R Meningitis/Encephalitis (ME) panel, have significantly reduced the time for detection of the viral etiology of PVM (188,189).…”
