Multivalency as a Key Factor for High Activity of Selective Supported Organocatalysts for the Baylis–Hillman Reaction

Abstract: The polystyrene-supported N-alkylimidazole-based dendritic catalysts for the Baylis-Hillman reaction exhibit one of the strongest beneficial effects of multivalent architecture ever reported for an organocatalyst. The yields in the model reaction of methyl vinyl ketone with p-nitrobenzaldehyde are more than tripled when a non-dendritic catalyst is replaced by a second- or third-generation analogue. Moreover, the reaction of the less active substrates will not occur with the non-dendritic catalyst and will proc… Show more

“…In fact, the catalytic activity of 3-FlC2-Pro-Tyr-Asp-Ado-NH 2 was found to be much lower (7% yield in 24 h, see ESI † ) than that of Fl-Pep5-b ( Table 1 , entry 17) in the sulfoxidation of thioanisole under the same conditions, showing the importance of the hydrophobic support resin that would make the flavopeptide conformationally profitable. 21 …”

“…Chemical catalysis is another mechanism by which polyvalent recognition events can be magnified in their effect. Although polyvalent catalysts have been intensively investigated for practical reasons in synthetic chemistry—for example, to increase activity by virtue of high local concentration and recyclability by virtue of easier recovery—they have only rarely been tested as a vehicle for information transfer in model systems …”
