Nano-Targeting of Mucosal Addressin Cell Adhesion Molecule-1 Identifies Bowel Inflammation Foci in Murine Model

Abstract: Anti-MAdCAM-1 functionalized nanoparticles detected active bowel inflammation foci, accurately following MAdCAM-1 expression pattern. These nanoconjugates could be a promising noninvasive imaging system for an early and accurate follow-up in patients affected by acute colitis.

“…Colony formation assay. colon cancer SW480 and T84 cells (1,000 cells/dish) were seeded in 3.5 cm dishes and cultured in a humidified environment with 5% CO 2 at 37˚C for 24 h. Then, different concentrations of scutellarein (10,20,40,60 and 80 µM) were added into each dish and the cells were incubated with scutellarein at 37˚C for 15 days. Cells treated with dMSO were used as the negative control.…”

“…The increased expression of McAM in the primary tumor types is associated with metastasis and prognosis in several cancer types (16)(17)(18). Mucosal addressin cell adhesion molecule 1 (Madcam1) is known as a type of adhesion molecule, which is involved in inflammatory responses with no expression in the majority of normal tissues (19,20). Receptor for advanced glycation end products (RAGE) is a signal transduction receptor that is involved in multiple pathologic conditions, including carcinogenesis and inflammation relying on its diverse ligands (21,22).…”

“…81 In another study, Truffi et al developed anti-MAdCAM-1 antibody-functionalized MnO nanoparticles with low toxicity. 82 By actively targeting the MAdCAM-1, the MnO nanoparticles achieved enhanced accumulation in the inflamed bowel sites in a murine model of acute colitis.…”

“…Imaging has long been utilized to evaluate segments of the digestive tract that are inaccessible with conventional endoscopy. For decades, nanoparticles such as Au, 116,117 Fe and Au complexes, 118 quantum dots, 119,120 MnO, 82 and lipid nanoparticles 74 have been developed and used in cancer imaging and diagnosis. During traditional clinical CT imaging, iodinated small molecules and barium sulfate suspensions are usually used as contrast agents, 121,122 but they are not ideal for imaging IBD due to their nonspecificity.…”

“…Due to their size and versatility, nanoparticles have attracted interest in the field of anticancer medicine, and they have shown ability to deliver consistent amounts of drugs and control their release at the tumor site [14,15,16,17,18]. Interesting feature of nanoparticles and nano-systems is the possibility to chemically or genetically modify their surface with a variety of targeting moieties or active ligands in order to trigger specific direction and recognition of the biological target [19,20,21]. The aim of the present paper is to review the current state-of-the-art and future translational perspectives on nano-strategies to target breast cancer microenvironment.…”
