Nanoparticle-Based Drug Delivery System—A Target Strategy for Osteoarthritis Treatment

Abstract: Osteoarthritis (OA) is a bone and joint disease with pathological characteristics such as articular cartilage degeneration injury and synovial and subchondral bone reactive hyperplasia. Once cartilage is damaged, it is difficult to repair it by itself. Current clinical practice is mainly limited to symptomatic treatment, not changing the degenerative process of osteoarthritis. The important goal of nanomedicine is targeted delivery. Nanoparticles (NPs) can provide many unique potential functions for the target… Show more

“…29 Although these methods demonstrate certain therapeutic effects, they also possess drawbacks such as donor site damage, inconsistency between the repaired area and surrounding cartilage characteristics, and poor interface healing. 30 Consequently, enhancing the implementation of clinical guidelines, including weight loss and rehabilitation programs, as well as effective pain control, was considered a crucial aspect for improving the quality of life and functionality of individuals with OA. 6 In our research, static and dynamic behavioral analysis has been widely recognized as a valuable reference for assessing pain control, quality of life, and functional abilities in individuals with OA.…”

“…As a transportation system for bioactive factors, a scaffold can ensure their stable and sustained release, preserve their biological activity and simulate the natural bone microenvironment [ 19 ]. In recent years, there has been strong interest in bone-targeted therapy based on scaffold materials due to the increasing number of patients requiring pharmacological treatment for bone tissue disorders [ 26 , 27 ]. Indeed, n-HAp composite scaffolds can become a promising option as a targeted delivery system based on bone tissue engineering scaffold and possess superiority to n-HAp, involving great osteoconductivity and supporting cellular responses for bone reconstruction, as well as upgrading the poor mechanical property of n-HAp [ 23 , 28 ].…”

“…As a degenerative disease characterized by cartilage damage with chronic pain and functional impairment, osteoarthritis (OA) not only reduces the quality of life of patients, but also leads to a growing social health care burden [ 1 , 2 ]. Although painkiller and artificial joint replacement are available for symptomatic relief of advanced OA, therapies to cure the disease radically are still lacking [ 3 , 4 ]. Therefore, early diagnosis and intervention of OA is particularly important to slow down the progression of the disease, improve quality of life in patients with OA.…”

“…The nanoplatform provides opportunity for improved relaxivity of contrast agents, enhancement of specific distribution, and maintenance of effective concentration [ 3 , 4 ]. Size of nanoparticles is a principal factor for the ability to pass through gliding surface and enter the articular cartilage, for superficial zone do not covered by perichondrium but is a structure formed by dense collagen fibers arranged in parallel, the gaps of which is around 60 nm [ 12 ].…”
