Natural killer cell immunotherapy against multiple myeloma: Progress and possibilities

Abstract: Multiple myeloma (MM) is a complex aggressive mature B-cell malignancy. Although with the wide application of chemotherapy drugs, it remains incurable and the vast majority of patients relapse. Natural killer (NK) cells, also known as CD56 CD3 large granular lymphocytes, are cytotoxic innate immune cells against MM without prior sensitization steps. NK cell-based immunotherapy is extensively promising in a wide range of clinical settings. It is worthy of note that some novel drugs such as monoclonal antibodies… Show more

“…This finding reveals that NK cells are the main secretors of GzB in normal conditions and have the main role in the body defence mechanism against cancer. Moreover, these cells have the ability to attack and decompose the foreign bodies and can be used in developing a new treatment based on immunotherapy as reported by Liu Jin et al (Liu et al 2018) As for cytokines, expression levels of IL-1 were increased 14000 fold more than the healthy control in the plasma. This is consistent with recent studies that analyzed the expression of IL-1 in the tumor microenvironment and reported that IL-1 has a role in tumor progression.…”

“…NK cells kill MM cells by releasing lytic granules containing granzymes and perforin [99,100]. NK cells recognize myeloma cells that downregulate the MHC I receptor and express high levels of MICA [52,101,102].…”

“…Each year 3-5 cases per 100,000 people are diagnosed (12). The disease is primarily characterized by anemia, increased risk of infection, hypercalcemia, bone lesions and renal failure and it is the second most common hematological malignancy (13). MM interferes with the mechanisms of bone renewal causing its decalcification (osteolytic bone lesions) that usually cause intense bone pain and increase the risk of spontaneous fractures.…”
