Background: About 80% to 85% of lung cancers are non-small cell lung cancer (NSCLC), which are believed to secrete mediators that may lead to disorders in the immune system and tumor immune surveillance. This disorder may include dysfunction in CD8 + T cells and NK cells, which play critical role in fighting against cancer. Aim: Evaluate the impact of lung cancer on the numbers and function of CD8 + T cells, NK and NK T cells as well as the inflammatory milieu. Materials and Methods: Peripheral blood samples were collected from healthy volunteers (n=5) and early diagnosed lung cancer patients (NSCLCs; n=5). The numbers of CD8 + T cells, NK cells and NK T cells were analyzed by flow cytometry after staining with the relevant antibodies. Plasma was prepared from the blood to measure the proinflammatory cytokines and chemokines by Lumiex® protein array. Results: Compared to healthy controls, lung cancer patients showed significant decrease in numbers of CD8 + T cells and NK cells. NK cells showed considerable reduction in their expression levels of the cytolytic molecule granzyme-B. Marked higher levels of IL-1 and CCL2 was detected in the plasma of lung cancer patients as compared to healthy controls. Conclusion: Early diagnosed lung cancer patients showed dysfunction not only in the numbers of cytolytic immune cells but also their function that correlated with inflammatory microenvironment.

Multiple myeloma is an incurable cancer characterized by the uncontrolled growth of malignant plasma cells nurtured within a permissive bone marrow microenvironment. While patients mount numerous adaptive immune responses directed against their disease, emerging data demonstrate that tumor intrinsic and extrinsic mechanisms allow myeloma cells to subvert host immunosurveillance and resist current therapeutic strategies. Myeloma downregulates antigens recognized by cellular immunity and modulates the bone marrow microenvironment to promote uncontrolled tumor proliferation, apoptotic resistance, and further hamper anti-tumor immunity. Additional resistance often develops after an initial clinical response to small molecules, immune-targeting antibodies, immune checkpoint blockade or cellular immunotherapy. Profound quantitative and qualitative dysfunction of numerous immune effector cell types that confer anti-myeloma immunity further supports myelomagenesis, disease progression and the emergence of drug resistance. Identification of tumor intrinsic and extrinsic resistance mechanisms may direct the design of rationally-designed drug combinations that prevent or overcome drug resistance to improve patient survival. Here, we summarize various mechanisms of immune escape as a means to inform novel strategies that may restore and improve host anti-myeloma immunity.

Background: Previous reports have been analyzed the prevalence/association of apical periodontitis (AP) with systemic diseases. The present study aims to analyze the prevalence of healthy/diseased periapex and endodontic treatments in patients with Multiple Myeloma (MM) and compare the results with those of control subjects. Material and Methods: Panoramic radiographs of 50 individuals with MM were evaluated and compared with 50 controls that were sex and age matched exactly with the diseased group. Radiographic analysis was performed by two experienced endodontists under standardized conditions. The periapical status (presence or not of AP) was assessed using the periapical index (PAI). Data included systemic health, technical quality of root fillings, total number of teeth, quality of restoration, and periapical status. Statistical evaluation of differences between groups used chi-squared tests and Fisher's exact tests. Results: The prevalence of root canal-treated teeth was 10.11% in the MM group and 12.05% in the control group (p=0.90). The average root canal-treated teeth in the test group was 2,34 and 2.48 in the control group, where the difference was statistically significant (p=0.05). AP in 1 or more teeth was found in 86 % and in 78% of the patients in the MM and the control groups, respectively. When analyzed by subject, there was no statistically significant difference in the prevalence of AP (p>0.72). Similarly there was also no statistically significant difference in the prevalence of PA (p=0.85), when analyzed by tooth, AP was found in 63.2% and 62.9% in MM and control groups. Conclusions: The presence of AP and endodontic treatment was not significantly different in individuals with MM compared with control subjects. Future studies are needed to elucidate and confirm the association between MM and AP.
