Natural Kinase Inhibitors for the Treatment and Management of Endometrial/Uterine Cancer: Preclinical to Clinical Studies

Abstract: Endometrial cancer (EC) is the sixth most prevalent type of cancer among women. Kinases, enzymes mediating the transfer of adenosine triphosphate (ATP) in several signaling pathways, play a significant role in carcinogenesis and cancer cells’ survival and proliferation. Cyclin-dependent kinases (CDKs) are involved in EC pathogenesis; therefore, CDK inhibitors (CDKin) have a noteworthy therapeutic potential in this type of cancer, particularly in EC type 1. Natural compounds have been used for decades in the tr… Show more

“…Parameters set for 50 trajectory frames within the time span of 10 ns. Distance restraints with global weight of 1.0, and Poisson-Boltzmann/generalized born (PB/GB) molecular mechanics were considered for conformational stability of the PIK3CA-streptenol derivative complex system [ 33 , 34 ].…”

“…Chemical modification of natural product molecules has also yielded multiple clinically relevant drugs such as caspofungin (Walsh et al, 2004), artemether (Hien et al, 1996), or etoposide (Cragg and Pezzuto, 2016;O'Dwyer et al, 1985). As a reflection of the high significance of natural products for the discovery and development of new pharmaceuticals, a high number of natural products or their derivatives are currently in clinical trials for a range of diverse diseases (Ahmad et al, 2021;Lima et al, 2021;Nile and Kai, 2020;Singla et al, 2022aSingla et al, , 2022b. Moreover, multi-component natural product mixtures are also studied to explore their therapeutic potential and used clinically (Wink, 2015).…”

“…For example, paclitaxel, camptothecin, homoharringtonine, ginsenoside Rg3, etc. In recent years, with the deepening of understanding and attention to natural products, many new opportunities have been brought to the development of natural anti-tumor molecules ( Singla et al, 2021a ; Singla et al, 2021b ; Tan et al, 2021 ; Wang et al, 2021 ; Singla et al, 2022 ).…”
