Nerve Growth Factor Treatment Prevents the Increase in Superoxide Produced by Epidermal Growth Factor in PC12 Cells

Abstract: Stimulation of pheochromocytoma (PC12) cells with the mitogen epidermal growth factor (EGF) produced a rapid and robust accumulation of intracellular reactive oxygen species (ROS), an accumulation which, in other systems, has been shown to be essential for mitogenesis. Brief pretreatment of the cells with nerve growth factor (NGF) suppressed the EGF-mediated ROS increase. EGF failed to produce elevations in ROS in a PC12 variant stably expressing a dominant-negative p21ras construct (PC12-N17) or in cells pret… Show more

“…This idea is in agreement with recent reports on redox signaling by NGF receptors in neuronal cells (45,46). Although not biochemically characterized in detail, these oxidant species include probably both hydrogen peroxide and superoxide, based on the data obtained with fluorescent redox probes with different radical specificity (DCF-DA and DHE).…”

“…Several experimental results indicate that signal transduction mediated by growth factors is accompanied by transient accumulation of ROS and that, in the absence of growth factors, exposure of cells to H 2 O 2 mimics some molecular events that follow mitogenic stimulation, including Tyr phosphorylation of growth factor receptors (GF-Rs) and the activation of downstream cascades (1,11). Diethylmaleate (DEM) is an indirect oxidant that yields an accumulation of endogenously produced ROS through a GSH depletion (20 and DEM induce ERKs activation.…”
