Neuregulin 1 improves complex 2-mediated mitochondrial respiration in skeletal muscle of healthy and diabetic mice

Abstract: It has been reported that neuregulin1 (NRG1) improves glucose tolerance in healthy and diabetic rodents. In vitro studies also suggest that NRG1 regulates myocyte oxidative capacity. To confirm this observation in vivo, we evaluated the effect on mitochondrial function of an 8-week treatment with NRG1 in db/db diabetic mice and C57BL/6JRJ healthy controls. NRG1 treatment improved complex 2-mediated mitochondrial respiration in the gastrocnemius of both control and diabetic mice and increased mitochondrial comp… Show more

“…NRG1 regulates myogenesis 9 , muscle spindle development and maintenance 10 , glucose transport 11 , 12 , mitochondrial oxidative capacity 13 , 14 and acetylcholine receptor expression 8 , 15 . A role of the NRG1/ErbB system in muscle regeneration has been postulated, and NRG1 expression increases in satellite cells and motoneurons after toxin-induced muscle damage 16 .…”

“…Previous studies revealed, NRG1/ErbB pathway is impaired in DCM cells which suggested NRG1/ErbB pathway may play an important role in DCM (30)(31)(32). Besides, NRG1/ErbB can improve glucose tolerance in healthy and diabetic rodents (32). And, also previous studies showed NRG1 can regulate myocyte oxidative capacity (33,34).…”

“…Therefore, inhibiting oxidative stress is believed to be an important strategy for the therapy of DCM. Besides, NRG1/ErbB can improve glucose tolerance in healthy and diabetic rodents (32).…”

“…While circulating NRG1 has previously been shown to act directly on the central nervous system [22,55] , liver [36,55] , skeletal muscle [56] , and cardiac muscle [24] here we propose an additional site of action within adipose tissues. We present a model that Type III NRG1 is epigenetically regulated and endogenously produced within adipose as a molecular rheostat controlling adipose expandability.…”
