Neurokinin‐1 receptor localisation in guinea pig autonomic ganglia

Abstract: We have used multiple-labelling immunohistochemistry and confocal microscopy to determine the distribution of immunoreactivity to the tachykinin neurokinin-1 (NK(1)) receptors in guinea pig sympathetic ganglia. Although nerve fibres containing immunoreactivity to substance P were common in all ganglia except the superior cervical ganglia, most neurons expressing NK(1) receptor immunoreactivity were not closely surrounded by pericellular baskets of substance P-immunoreactive boutons. Conversely, many neurons su… Show more

“…However, the excitability of some vasomotor neurones can be enhanced by peptides released from collaterals of unmyelinated sensory neurones, most of which are likely to be visceral nociceptors. Whilst collaterals of unmyelinated nociceptors are rare in the paravertebral sympathetic ganglia, they are abundant in the prevertebral ganglia (Matthews et al 1987), where they ramify extensively between vasomotor neurones and neurones regulating gastrointestinal motility and secretion (Messenger et al 1999). These fibres project into the pre-vertebral ganglia from the mesenteric circulation and the gut (Kreulen & Peters 1986.…”

“…This variant lacks the intracellular C-terminus of the full-length NK1 receptor and is not desensitized following agonist binding. Furthermore, it inhibits the channels responsible for the depolarization and LAH via an intracellular signalling pathway that is completely unknown (Messenger et al 1999.…”
