Neuromuscular Blocking Action of Alkaloids from a Japanese Crude Drug “Shin-I” (Flos Magnoliae) in Frog Skeletal Muscle

Abstract: Fromthe dried buds of Magnolia salicifolia MAXIM, d-coclaurine (1), d-reticuline (2), 1-N-methylcoclaurifle (3) and yuzirine (4) were isolated. Fifty percent inhibitory doses (1iM) in acetyicholine (ACh, 27.5 tM)-induced contraction of frog skeletal muscle were as follows: d-tubocuraine (d-TC) 3.09, 2 93.2, 3211, 1 467 and 4 more than 945. 4 (94.5 and 315 tM) increased the ACh-induced contraction. Indirectly stimulated twitching of frog nerve-muscle preparations were completely blocked by the following doses (… Show more

“…It has long been used for treatment of headaches and nasal diseases, as a tranquilizer and analgesic, and is considered to be especially efficacious for empyem, chronic inflammation of paranasal sinuses. It has been pharmacologically found to act as an inhibitor on the central nervous system (1,2), to effect neuro-muscular blocking (3,4), and to possess anti-allergic effects (5).…”

“…A total alkaloidal extract from the aerial parts of L. cubeba showed antimicrobial activity against Staphylococcus aureus. Further purification of this extract led to the isolation of the new compounds 1 and 2, together with the 11 known analogues lirioferine (3) [10], isoboldine (4) [11], boldine (5) [12], norlirioferine (6) [13], laurelliptine (7) [14], wilsonirine (8) [10], N-(methoxycarbonyl)-N-norisoboldine (9) [15], isocorydine (10) [12], muricinine (11) [16], reticuline (12) [10], and N-methylcoclaurine (13) [17]. The structural elucidations of all compounds were based on spectroscopic evidence and comparison with data reported in the literature.…”

“…Several active constituents exhibiting various biological activities from Magnolia Flower have been identified, according to the literature, including lignans with IL-2 inhibitory 19) and cholesterol-lowering 20) activity, neolignans with antiinflammatory 21) and anti-vasculogenesis 22) activity, and volatile oils with anti-inflammatory activity. 23) There have also been many reports on the isolation of volatile oils, 24) lignans, 25) neolignans, 25) phenylpropanoids, 26) alkaloids, 27) monoterpene glycosides, 28) and flavonoids 15) from Magnolia species in efforts to study biological activity.…”
