The impact of peripheral nerve damage on a patient's quality of life is severe. The most frequent peripheral nerve crush damage is a sciatic nerve injury. Previous research has shown that glibenclamide (GB) has neuroprotective properties in a variety of oxidative stress‐related disorders, including Alzheimer and Parkinson. The goal of this study was to see how GB affected nerve regeneration and improved function of the sciatic nerve in a rat model following a crush injury. We evaluated motor function, sensory recovery, gene expression, and histomorphometry following damage at different time points. Additionally, we assessed atrophy in the gastrocnemius muscle using histology and mass ratio analyses. Our results suggest that 2, 4, 6, and 8 weeks following glibenclamide therapy, promotes the recovery of motor and sensory function in the injured site. Following glibenclamid injection, the mRNA levels of neurotrophic factors (NGF and BDNF) are raised. According to histomorphometry assessment, glibenclamide injection also increased the number of myelinated fibers while decreasing their thickness. These results showed that glibenclamide therapy by decreasing the proinflammatory and oxidant factors may enhance the nerve regeneration. It is clear that more research is needed to confirm these findings.

“…Since the combination of lovastatin and AICAR results in progress of protective Th2 immune response, it is concluded that this combination could be one of the approaches for the treatment of CNS inflammation and neurodegeneration in MS. By summarizing all arguments which were mentioned above, it can be concluded that lovastatin in combination with RLP and AICAR are known to be promising and efficient approaches for the treatment of MS introducing minimal adverse and toxic effects. Moreover, other studies suggest that lovastatin can be used as a therapy for other neurological disorders such as Alzheimer's disease, Early Stage Parkinson's Disease and other peripheral nerve injuries that works with the same mechanism which was discussed above [69][70][71][72][73][74]. There is evidence that statins which are mainly used in treatments of dyslipidemia can be used for attenuating symptoms of cardiovascular and neurodegenerative diseases. Lovastatin and other statins are known to function through inhibition of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase which is involved in the majority processes such as cell differentiation, proliferation and migration. Recently it was revealed that lovastatin is effective in the reducing total and low-density-lipoprotein cholesterol, lowering the risks of post-surgical complications and mitigation of anticancer drugs’ side effects. Moreover, there are other combinations with other drugs to treat cardiovascular diseases. In addition, antiinflammatory and immunomodulatory effects of lovastatin diminish neurological disorders such as multiple sclerosis (MS). Especially, combination treatment of lovastatin and rolipram in suboptimal doses is considered to be the most promising approach for protecting neuronal axons from demyelination and promoting neuro repair in MS. Although lovastatin demonstrates promising option for treatment of cardiovascular and neurological diseases, more clinical trials and studies in vivo and in vitro are required.

Background: Peripheral nervous system injuries are common and currently have no definitive treatment method. Phenytoin is one of the main antiepileptic drugs. Some studies have described a cerebroprotective effect of phenytoin in an established model of global cerebral ischemia. Objectives: In this study, the neuroprotective effects of phenytoin were evaluated on the cultivation and maintenance of Wharton’s jelly stem cells (WJSCs) on acellularized sciatic nerve scaffolds. Methods: In this study, acellular scaffolds from the rat sciatic nerve were prepared by the sondell method. After extraction of cells of MSCs, flow cytometry analysis was executed. Also, cell differentiation potential was evaluated by placement in osteogenic and adipogenic differentiation media for 21 days. Biocompatibility of the scaffold and cell viability were investigated using the MTT assay. The morphological and cell adhesion characteristics of MSCs on acellular scaffolds were compared using SEM micrographs images. Data were analyzed using the one-way analysis of variance (ANOVA) and Tukey post hoc test by SPSS (version 19.0) software. Results: The removal of cells from the scaffold was confirmed by stanning with hematoxylin-eosin, van Gieson's picro-fuchsin, and DAPI. With the aid of flow cytometry analysis and differentiation into bone and fat cells, it was confirmed that extracted cells were mesenchymal stem cells. The results of the MTT assay showed that phenytoin increased cell viability and retention on the scaffold. Conclusions: The study indicated that phenytoin improves the viability of cells and provided a good condition for the growth, survival, and attachment of cells to the scaffold when compared to the control group. These results suggest that phenytoin can be considered a new treatment for nerve regeneration and tissue engineering applications.
