New drugs for methicillin-resistant Staphylococcus aureus skin and soft tissue infections

Abstract: Purpose of reviewStaphylococcus aureus is a pathogen incriminated in skin and soft tissue infections (SSTIs), with methicillinresistant S. aureus (MRSA) becoming the predominant cause and representing a significant burden to the healthcare system. The last updated Infectious Diseases Society of America (IDSA) guidelines concerning MRSA infections and SSTIs management were published in 2011 and 2014, respectively. The UK updated guidelines for MRSA infection treatment were published in 2021. Older treatment opt… Show more

“…Infection at the surgical site (SSI) is considered one of the most important complications after surgery. When infection occurs after surgery, the use of long-term antibiotics is necessary to control the infection, which increases the patient's hospitalization and increases the cost of medical care (17). Treatment of MRSA infections has become challenging due to resistance to a wide range of beta-lactam antibiotics.…”

“…Several new antibiotics with good gram-positive activity were made available over the last 2 decades to treat skin and soft tissue infections [ 28 ]. These include delafloxacin, omadacycline, tedizolid, dalbavancin, telavancin, oritavancin, and eravacycline, which are available in either oral or IV formulation.…”

“…As such, the use of modified glycopeptide derivatives such as dalbavancin, oritavancin, and telavancin, and/or combinational antibiotic therapy is typically exercised as a way to overcome AMR [ 485 ]. While there remains a limited supply of novel antibiotics in development [ 486 , 487 , 488 , 489 ], the emergence of resistance to future approved antibiotic treatment regimens is expected [ 490 ]. Although R&D pathways surrounding new-class antibiotics represent a possible path forward, these programs remain high risk, expensive, and time-consuming endeavors that many pharmaceutical companies have withdrawn from [ 9 , 491 , 492 , 493 ].…”
