New Micropeptins with Anti-Neuroinflammatory Activity Isolated from a Cyanobacterial Bloom

Abstract: Metabolite mining of environmentally collected aquatic and marine microbiomes offers a platform for the discovery of new therapeutic lead molecules. Combining a prefractionated chromatography library with liquid chromatography tandem mass spectrometry (LC-MS/MS)-based molecular networking and biological assays, we isolated and characterized two new micropeptins ( 1 and 2 ) along with the previously characterized micropeptin 996. These metabolites showed potency in … Show more

“…The material in this library was derived from a cyanobacterial bloom in Muskegon, MI . This library was used by our laboratory previously to isolate new antineuroinflammatory micropeptins . In addition, to these antineuroinflammatories there were also chromatography fractions that showed strong cytotoxicity to murine BV-2 microglial cells .…”

“…Finally, photosynthetic prokaryotes are still largely unexplored biomanufacturing platforms, despite producing a vast repertoire of unique compounds [338], including molecules with antioxidant and anti-inflammatory properties [339,340] like the pigment-protein complex phycocyanin, which is a selective inhibitor of pro-inflammatory oxylipin synthesis [341,342], and various polysaccharides [343][344][345] and peptides [346][347][348]. Last but not least, an increasing number of genetic engineering and synthetic biology tools [349] can be employed to produce recombinant therapeutics in cyanoprokaryotes [350], as already reported for immunomodulatory proteins [351].…”

“…Tandem mass spectrometry (LC-MS/MS) was performed using a ThermoFisher Scientific LTQ XL linear ion trap mass spectrometer with an electrospray ionization (ESI) source. Analytical HPLC was carried out using a Dionex UltiMate 3000 HPLC system equipped with a micro vacuum degasser, an autosampler, and a diode-array detector [25]. Samples were separated on a Phenomenex Luna C 18 column (5 µm; 150 × 4.6 mm).…”
