NHA2 is expressed in distal nephron and regulated by dietary sodium

Abstract: Increased renal reabsorption of sodium is a significant risk factor in hypertension. An established clinical marker for essential hypertension is elevated sodium lithium countertransport (SLC) activity. NHA2 is a newly identified Na+(Li+)/H+ antiporter with potential genetic links to hypertension, which has been shown to mediate SLC activity and H+-coupled Na+(Li+) efflux in kidney-derived MDCK cells. To evaluate a putative role in sodium homeostasis, we determined the effect of dietary salt on NHA2. In murine… Show more

“…The same study demonstrated functional coupling of NHA2 to the V-type H + -ATPase at the plasma membrane in the renal MDCK cell line to create a virtual Na + efflux pump, and suggested that NHA2 functionally recapitulates the ancient bacterial NhaA. Endogenous NHA2 was also described recently in the renal distal convoluted and connecting tubules where it was upregulated in response to a high-Na + diet (156), a physiological response consistent with its contribution to Na + efflux rather than reabsorption. The cellular localization of NHA2 and directionality of NHA2-mediated Na + transport in the mammalian intestinal epithelia is not known.…”

“…; Kondapalli et al . ). NHA2 expression in kidney is confined to the distal nephron and renal collecting duct, regions that play critical roles in salt, pH and volume homeostasis (Fuster et al .…”

“…Principal cells maintain sodium and water balance and the intercalated cells control acid–base homeostasis (Kondapalli et al . ). Although the physiological function of NHA2 is largely obscure, defining the pathways regulating NHA2 expression is an important first step towards deciphering its potential role in hypertension and kidney disease.…”

“…NHA2 has been proposed to function as a unique cell acid loader in MDCK cells that electrically exchanges 2 extracellular H + for 1 intracellular Na + /Li + , although transport direction depends on transport stoichiometry and ion gradients (Kondapalli et al . ). NHA2 appears electroneutral in other preparations.…”

“…Kondapalli et al . () localized NHA2 to the apical membrane of the distal tubule where it would reduce or reverse net transepithelial Na + reabsorption as an acid loader, but promote Na + hyperabsorption as an acid extruder. Further studies are essential for clarification.…”
