NK Cell Memory to Cytomegalovirus: Implications for Vaccine Development

Abstract: Natural killer (NK) cells are innate lymphoid cells that recognize and eliminate virally-infected and cancerous cells. Members of the innate immune system are not usually considered to mediate immune memory, but over the past decade evidence has emerged that NK cells can do this in several contexts. Of these, the best understood and most widely accepted is the response to cytomegaloviruses, with strong evidence for memory to murine cytomegalovirus (MCMV) and several lines of evidence suggesting that the same i… Show more

“…It is also important to consider factors other than simple presence of absence of genes which could impact KIR expression. These could include NK developmental processes that are influenced by genetic factors outside the KIR locus, such as the influence of HLA on NK cell education (44) or HCMV seropositivity, which is known to increase KIR expression on peripheral NK cells (45). HCMV seropositivity could also be relevant because uNK can respond to HCMV-infected stromal and trophoblast cells in a manner that is at least partially mediated by KIR2DS1 (46), highlighting a potential for KIR2DS1 expression to protect against HCMV-mediated early pregnancy failures.…”

“…However, HCMV has developed strategies to effectively escape recognition by NK cells and prevent NK cell killing through virus-encoded gene products (18,19,24), such as UL18, UL40, UL16, UL141, UL142, and UL83, which inhibit the CD155 ligand presentation for the NK cell activating receptor NKG2D on infected cells (25,26). Therefore, when live-attenuated or disabled viral vaccine platforms are employed, deleting these genes that inhibit NK cell killing on CMV-infected cells could be relevant in controlling infection (27).…”

“…It has been shown that following mouse-cytomegalovirus (MCMV) virus infection, NK cells can show long-lived selfrenewing antigen-specific memory phenotype and respond to recall response. 21,22 NK cell memory response is discussed elsewhere [23][24][25] and beyond the scope of this review.…”
