Non-canonical WNT/PCP signalling in cancer: Fzd6 takes centre stage

Abstract: Frizzled receptors are the mediators of the wnt canonical and non-canonical pathways, which play fundamental roles in cell differentiation and organism development. A large body of work indicates that dysregulation of wnt signalling is a feature of oncogenic transformation, but most of the studies published so far focus on the assessment of the consequences of aberrations of the canonical pathway in human cancer. In this review, we discuss the emerging role of the wnt non-canonical pathway regulated by frizzle… Show more

“…5c). Wnt5a is an important regulator of both the Wnt/PCP and Wnt/β-catenin pathways, playing a prominent role in neuronal stem cells and nervous system development 67,68 , and is an important regulator of tumorigenicity of many human cancers, including neuronal derived ones 62,6772 .

“…Corda et al . reported a genomic amplification of FZD6 in breast cancer 41 , 42 and Cantilena et al . pointed that FZD6 could mark highly tumourigenic stem-like cells in mouse and human neuroblastomas 43 .…”

“…The Fzd6 (frizzled class receptor 6) gene is a member of the ‘frizzled’ gene family, which encode 7-transmembrane domain proteins that are receptors for Wnt signaling proteins. FDZ6 has a known role in non-canonical WNT/PCP signalling in cancer (Corda and Sala 2017), including mediating transformation, increased invasiveness of tumour cells and metastasis (Cantilena et al 2011; Corda and Sala 2017; Corda et al 2017). In agreement with this, increased expression of FDZ6 has reported in many cancer types, and correlates with poor prognosis in patients with breast, brain and oesophageal cancer (Corda et al 2017; Huang et al 2016; Zhang et al 2019).…”
