Normal-phase high-performance liquid chromatographic separation of procyanidins from cacao beans and grape seeds

Abstract: An HPLC method using a normal-phase silica column and a gradient of dichloromethane-methanol-formic acid-water mixtures as the eluent was developed to separate procyanidins on a molecular mass basis, without derivatization. It was successfully applied to the analysis of procyanidin extracts from cacao beans and grape seeds. The monomers and major dimers were resolved as discrete peaks. Oligomeric and polymeric components were eluted in order of increasing degree of polymerization, as confirmed by determining t… Show more

“…Amount and composition. The quantity of extracted procyanidins per berry was determined by acid-catalyzed degradation in the presence of benzyl mercaptan (thiolysis) (Rigaud et al, 1993;Matthews et al, 1997). The quantities of extracted material declined from a high value at ve raison ( Fig.…”

“…The building blocks of PAs are flavan-3-ol oligomers which have 3 rings (ring A: triketide, ring B: phenylpropanoid, ring C: pyran which is formed by condensation). The main flavan-3-ol units in grape seed extract (GSE) PA are catechin (C), epicatechin (EC), catechingallate (CG) and epicatechingallate (ECG) 21) whilst the main monomeric flavan-3-ol unit in cocoa seed extract (COE) is EC unit 22) (Fig. 1).…”

“…HILIC is an aqueous variant of normal phase LC (NP-LC), which makes use of polar stationary phases (such as silica, bonded amide and diol phases) in Stellenbosch University http://scholar.sun.ac.za combination with apolar mobile phases. Compounds are separated based on polarity [84]. The added benefits of HILIC include its suitability for polar compounds not sufficiently retained in RP-LC, excellent compatibility with UV and MS detection, and it also allows for hyphenation to other modes of HPLC in 2-dimensional LC.…”
