Novel Concepts for HIV Vaccine Vector Design

Abstract: The unprecedented challenges of developing effective vaccines against intracellular pathogens such as HIV, malaria, and tuberculosis have resulted in more rational approaches to vaccine development. Apart from the recent advances in the design and selection of improved epitopes and adjuvants, there are also ongoing efforts to optimize delivery platforms.

“…The CanSino and Sputnik V vaccines specifically use Ad5, which is the same vector platform used in our studies. Ad5 is among the most well-studied vaccine vectors due, in part, to its extraordinary immunogenicity ( 2 , 26 ). Although SARS-CoV-2 vaccines prevent severe disease and death, they do not always confer durable sterilizing immunity, warranting the further optimization of current vaccine regimens.…”

“…Viral vectors are the best delivery tools for vaccine development, because they have intrinsic adjuvant capability and unique cellular tropism, and they are able to trigger robust adaptive immune responses. A number of new vectors, including Ad26 (adenovirus serotype 26), Ad35, poxvirus (e.g., canarypox virus-based ALVAC vector), replication competent Ad4, and CMV vectors, are currently being developed ( Table 2 ) [ 120 , 121 , 122 , 123 , 124 ]. However, viral vector vaccine platforms do have some limitations, such as preexisting immunity.…”
