Novel Fibroblast-Specific Monoclonal Antibodies: Properties and Specificities

Abstract: Specific detection of fibroblasts has been one of the unsolved problems in cell biology. Because monoclonal antibodies (MoAbs) might provide an easy and reproducible method of fibroblast detection, we have produced a panel of MoAbs raised against cell surface proteins of human dermal fibroblasts. Using flow cytometry and immunohistochemistry, we have shown that two of these MoAbs, FibAS01 and FibAS02, react exclusively with human fibroblasts. They do not react in vitro with human keratinocytes, endothelial cel… Show more

“…All cells also showed strong staining for other mesenchymal Ags, including vimentin, fibronectin, and collagen type I and III. Homogeneous cell surface expression of Ag ASO2, specific for fibroblasts (9), was also detected by flow cytometry, whereas Ag CD31, characteristic of endothelial cells, was missing.…”

“…Frozen tissue sections of human atopic eczema lesions were stained with an anti-CD1a biotin Ab to identify DC. Fibroblasts were identified using the anti-Thy-1 Ab AS02 (17,19,20,22). A close colocalization of CD1a-positive DC (brown) and Thy-1-positive fibroblasts (red) was found in the dermis of atopic eczema lesions (Fig.…”

“…The isolated proteins were separated by SDS-PAGE and blotted onto nitrocellulose membranes. The anti-Thy-1 mAbs or an isotype control IgG antibody followed by a secondary goat-anti-mouse antibody conjugated with alkaline phosphatase were used to detect the isolated proteins as described previously (Saalbach et al, 1996).…”
