“…We have not detected any requirements for DNA sequences upstream of the pIX SP1 site or downstream of the pIX TATA box that are important to pIX gene expression. Our studies support those of Matsui (21), which showed that sequences between nt -70 and -45 as well as the TATA box were important to pIX gene expression in transient expression assays, although the identity of the SP1 site was not recognized in that previous work. However, we have not found any sequences internal to the pIX gene that affect pIX gene expression in vivo.…”

“…Once again, the peak with the highest propensity value is at À70 bp relative to the TSS. A TATA box has been reported at position À30 in this promoter, to which TBP binds and directs transcription (53). There is also some other, unidentified activating upstream element from À45 to À70 bp relative to the TSS.…”

“…The pIX and E21 genes, like the Elb gene, have an Spl-binding site consensus sequence (GC box; 29) located within 30 bp 5' of a TATA box (50). Each GC box mediates transcription (14,31), and at least the E21 site (14), like the Elb site (63), binds purified Spl (14,63). No other adenovirus promoters have this arrangement of adjacent GC and TATA boxes, which may constitute the target of a shared mechanism for sequential transcription activation.…”
