Nucleotide Sequence of Mouse L19 Ribosomal Protein cDNA Isolated in Screening withtreOncogene Probes

Abstract: A cDNA library was prepared from cytoplasmic poly(A)RNA from mouse NIH-3T3 cells carrying a transfected human tre oncogene. Screening with tre gene probes identified a tre cDNA clone 11-4 and a co-purifying weakly hybridizing cDNA clone 11-5. The 11-5-specific RNA was expressed in both nontransfected and tre-transfected NIH-3T3 cells, showing it is of mouse rather than tre gene origin. Its nucleotide sequence was 717 bp long and contained, starting from the first nucleotide, an open reading frame of 588 bp fol… Show more

“…One reason for the excitement over these enzymes, however, comes from their possible role as target-speci®c regulators. Many UBPs have been identi®ed in genetic screens whose mutant phenotypes or patterns of expression implicate cellular processes as diverse as eye development in fruit¯y (Fischer-Vize et al, 1992;Huang et al, 1995), transcriptional silencing (Moazed and Johnson, 1996), stability of the yeast MATa2 transcription factor (Papa and Hochstrasser, 1993), cytokine response (Zhu et al, 1996(Zhu et al, , 1997, and oncogenesis (Nakamura et al, 1992). Most recently, a member of the UCH family was implicated in long-term facilitation of neuronal synapses in Aplysia (Hegde et al, 1997).…”

“…Tre2 was originally isolated in multiple transfection-based screens for novel oncogenes, by virtue of its potent ability to transform mouse NIH 3T3 cells (6,7). It was subsequently shown to function as a ubiquitin-specific protease (USP) in vitro, although specific target proteins whose turnover may be modulated by Tre2 have not been identified (8).…”

“…The results of Aoyama et al (1993) (using Western blot) are similar to our findings that the lowmolecular-weight heat shock protein crystalline alpha B (CRYAB) is upregulated in 70% of anaplastic tumors but not in most GBM. The ubiquitin carboxyl-terminal hydroxylase TRE-2 (TL132) is an oncogene widely expressed in human cancer cells but not in human cells from normal tissue (Nakamura et al, 1992).…”
