Nucleotide sequence of yst, the Yersinia enterocolitica gene encoding the heat-stable enterotoxin, and prevalence of the gene among pathogenic and nonpathogenic yersiniae

Abstract: The gene encoding the heat-stable enterotoxin (yst) was cloned from the chromosome of Yersinia enterocolitica W1024 (serotype 0:9), and the nucleotide sequence was determined. The yst gene encodes a 71-amino-acid polypeptide. The C-terminal 30 amino acids of the predicted protein exactly correspond to the amino acid sequence of the toxin extracted from culture supernatants (T. Takao, N. Tominaga, and Y. Shimonishi, Biochem. Biophys. Res. Commun. 125:845-851, 1984). The N-terminal 18 amino acids have the proper… Show more

“…Yst is synthesized as a pre-protoxin of 71 amino acid residues. The 18 W-terrninal residues constitute a typical signal sequence and the 30 C-terminal residues represent the mature toxin (Takao eta!., 1984;Guzman-Verduzco and Kupersztoch, 1989;Delor et al, 1990). STI and Yst also i-eaemble guanylin.…”

“…The virulence of Y. enterocolitica is controlled by chromosomal (yst, inv) (12)(13)(14)(15) and plasmid-encoded genes (16,17). The pYV virulence plasmid directs production of the outer membrane protein YadA and secretion of at least 11 antihost proteins called Yops (17)(18)(19).…”

“…Yersinia enterocolitica also secrete a heat-stable enterotoxin, but its role in bacterial virulence is uncertain. 21 The organism also can be carried in the bloodstream by phagocytic cells to more distant sites.…”
