Nutrition and fasting mimicking diets in the prevention and treatment of autoimmune diseases and immunosenescence

Abstract: Complex and coordinated signals are necessary to initiate and sustain the activation, proliferation, and differentiation of lymphocytes. These signals, which are known to determine T-cell fate and function, also depend on the metabolic state of the organism. Recent studies indicate that both the type and levels of nutrients can influence the generation, survival and function of lymphocytes and therefore can affect several autoimmune diseases. Here, we review the dysregulation of lymphocytes during autoimmunity… Show more

“…Caloric restriction improves chronic inflammatory and autoimmune disorders, such as multiple sclerosis and rheumatoid arthritis (Choi et al, 2017;Piccio et al, 2008). Intriguingly, gene modules associated with "inflammation of joint" and "rheumatoid arthritis" were 25 reduced in monocytes isolated from fasting mice ( Figure S5A and S5B).…”

“…Accordingly, the recent diet westernization of developing countries has been associated with an increased prevalence of inflammatory or autoimmune disorders (Manzel et al, 2014). In contrast, reduced caloric intake is associated with improved outcomes for metabolic, autoimmune and inflammatory diseases, including NAFLD (Kani et al, 2017), T2DM , CVD (Wei et al,10 2017), multiple sclerosis (Choi et al, 2016;Jahromi et al, 2014), rheumatoid arthritis (Choi et al, 2017;Kjeldsen-Kragh et al, 1991), asthma (Johnson et al, 2007), psoriasis (Jensen et al, 2016), colitis (Shibolet et al, 2002), and sepsis (Starr et al, 2016;Wannemacher et al, 1979) and has been shown to prolong the lifespan of rodents, monkeys, and humans (Fontana et al, 2010;Picca et al, 2017). However, the molecular mechanisms by which caloric restriction 15 modulates systemic inflammation remain poorly understood.…”

“…Periodic fasting (PF) and fasting-mimicking diets (FMDs) have been effective in increasing healthy lifespan or as therapies in mouse models for a variety of diseases (Choi et al, 2017; Lee and Longo, 2016; Brandhorst et al, 2015). FMDs can reduce cancer incidence and aging-associated immunosuppression/immunosenescence, a process aided by hematopoietic stem-cell-based regeneration (Brandhorst et al, 2015; Cheng et al, 2014).…”

“…Finally, although the strategy of alleviating or actually reverting autoimmune disorders in humans by fasting mimicking diets is among the most promising new potential therapeutic approach for these pathologies , caution is still necessary, especially regarding the feasibility of food deprivation in our species.…”
